摘 要:硫酸盐还原菌广泛存在于土壤、海水、河水、地下管道和油气储层中。硫酸盐还原菌可将硫酸盐还原为硫化氢,腐蚀金属管线和污染环境。硫酸盐还原菌作用产生的硫化氢可在环境治理中沉淀重金属离子。硫酸盐还原菌与人类的生产或生活密切相关,研究硫酸盐还原菌的生长繁殖规律具有重要意义。生长曲线可定量反映细菌的生长繁殖的规律。常用绝迹稀释法、镜下计数法1和浊度法测量硫酸盐还原菌的生长曲线。绝迹稀释法简便、准确可靠,但工作量和试剂耗费大,花费时间长。镜下计数法简便、快速,但不易将死菌和活菌区分开。浊度法简便、快速,但同样不能将死菌和活菌区分开,还不能区分硫酸盐还原菌与水中的其它细菌(如铁细菌或腐生菌)和难以用于混浊水样。本文提出根据生长过程中溶液硫离子浓度的变化测定硫酸盐还原菌的生长曲线。本法与浊度法测量结果类似,说明方法可行。本法比绝迹稀释法简便、快速。本法不受死菌和其它细菌的干扰和可用于混浊水样,比镜下计数法和浊度法优越。sulfate-reducing bacteria(SRB) widely exist in soil.sea water,river water,underground pipelines and oil and gas reservoir.SRB can make sulfate reduce to hydrogen sulfide which corrodes to the metal pipeline and pollutes the environment.Hydrogen sulfide produced can precipitate heavy metal ions for environmental management.SRB is closely related to the production and life of mankind.Growth curve can quantitatively reflect the bacterial growth rule.Vanishing dilution method,microscopic counting method and turbidity method commonly are used for determination of SRB growth curve.Vanish dilution method is simple,accurate and reliable,but the workload and reagent consumption are larger,and it takes longer time.Microscopic counting method is simple,fast,but it is not easy to distinguish between dead and vital bacteria.Turbidity method is simple,rapid,but it is not easy to distinguish between dead and vital too and yet distinguish between SRB and other bacteria in the water(such as iron bacteria or saprophytic bacteria) and it is difficult to be used for turbid water samples.The determination method of SRB growth curve from sulfur concentration changes is presented in this paper.The experiment results of this method and turbidity method are similar,and illustration of this method is feasible.This method is simpler and faster than vanishing dilution method.This method is not affected by dead bacteria and other bacterial interference and can be used in turbid water samples,and it is superior to microscopic counting method and turbidity method.
