摘 要:目的筛选纯化泄浊除痹方总皂苷效果最佳的树脂以及考察分离条件,以期得到高纯度的总皂苷。方法采用静态吸附-解析筛选树脂类型,并对影响分离的各种因素进行系统研究,以比色法测定泄浊除痹方总皂苷含量为评价指标,进行工艺筛选。结果 D-101分离效果最好,其最佳工艺为药液皂苷浓度5.205 mg.mL-1,上样量为1.5BV(树脂床体积),以1.0 BV.h-1的吸附速率进行吸附,30%乙醇5 BV进行洗脱效果最佳。经D-101处理后的泄浊除痹方总皂苷可达80%以上。结论该方法简单可行,分离效果好,能满足大生产要求。Objective To study the best resins and the optimum separation conditions for obtaining high-purity total saponins from Xiezhuochubi decotion.Methods The best macroporous resin was selected by using static adsorption and desorption of total saponins,and the factors affecting the separation of total saponins were investigated.Results The macroporous resin D-101 not only presented the highest absorption capacity for saponins from the Xiezhuochubi decotion among the 6 resins studied,but also had a good desorption capacity.The optimum absorption conditions were as follows: the loading volume was 1.5 BV,absorption rate was 1.0 BV·h-1 and 30% ethanol could elute most of the saponins with the volume of 5 BV.The purity rate of saponins from the Xiezhuochubi decotion was up to 80% after the isolation and purification by macroporous resin D-101.Conclusion This method is simple and feasible and can meet the need of the large-scale production.
