摘 要:目的利用单侧输尿管梗阻(UUO)小鼠肾纤维化模型,采用尾静脉注射的方法整体转染CTGF-siRNA质粒抑制肾脏结缔组织生长因子(CTGF)的表达,观察敲低CTGF表达对肾脏间质纤维化的影响。方法制备UUO模型及活体CTGF基因转染;应用免疫组织化学染色法检测α-平滑肌肌动蛋白(α-SMA)的表达;Western blot检测CTGF及α-SMA蛋白的表达;半定量RT-PCR检测α-SMA mRNA的表达。结果转染CTGF-siRNA质粒的UUO小鼠CTGF及α-SMA的基因表达明显下降(P<0.05)。结论经小鼠尾静脉注射CTGF siRNA质粒能显著抑制小鼠肾CTGF和α-SMA基因表达并能有效抑制小鼠肾脏间质纤维化,提示CTGF siRNA有潜力成为防治肾脏间质纤维化的一种新策略。Objective To observe the effect of expression of knockdown connective tissue growth factor (CTGF) on renal interstitial fibrosis in mice with unilateral ureteral obstruction (UUO), through caudal vein injection to transfect CTGF siRNA plasmid to inhibit the expression of connective tissue growth factor in mice with UUO. Methods The mouse models with UUO were established, then the CTGF gene transfection in the living mice were performed. The expression of α-smooth muscle actin ( α-SMA ) was detected by immunohistostaining; the expressions of CTGF and α-SMA were tested by Western blot ; the expression of α-SMA mRNA was detected by semiquantitative RT-PCR. Results The gene expressions of CTGF and α-SMA were obviously decreased in UUO mice who were transfected with CTGF-siRNA plasmid ( P 〈 0.05 ). Conclusion The transfection of CTGF siRNA plasmid via caudal vein injection can inhibit obviously the gene expressions of CTGF and α-SMA in kidney of mice and can effectively inhibit renal interstitial fibrosis in mice, which suggests that it is possible for CTGF siRNA to become a new way of preventing and treating renal interstitial fibrosis.
