摘 要:目的探讨醋酸泼尼松对生长期大鼠骨代谢指标的影响及不同剂量原儿茶醛对醋酸泼尼松性骨质疏松症的防治作用。方法 60只无特定病原体(SPF)级大鼠随机分为4组:正常对照组(NC组),模型对照组(GC组),高剂量原儿茶醛组(GHP组),低剂量原儿茶醛组(GLP组)。NC组每天灌胃0.9%氯化钠溶液,余3组分别灌胃泼尼松3.5 mg·kg-1·d-1造模,GHP组给予原儿茶醛8 mg·kg-1·d-1;GLP组给予原儿茶醛2 mg·kg-1·d-1。结果 GC组大鼠骨物理指标、骨密度减少,重要脏器比,骨矿物质和有机质下降(P<0.05);高、低剂量原儿茶醛治疗后大鼠骨物理指标,骨密度,重要脏器比,骨矿物质和有机质较GC组显著增加(P<0.05),与NC组比较差异无统计学意义(P>0.05)。结论原儿茶醛能明显改善模型动物的骨物理指标,增加骨密度、骨矿物质和有机质,增强重要脏器比,从而有效防治醋酸泼尼松致骨质疏松的发生。Objective To observe the effects of prednisone acetate on bone biochemical indicators in rats at growing period and to investigate the protective effect of different doses of protocatechualdehyde on osteoporosis induced by prednisone acetate in rats. Methods 60 Sprague-Dawley (SD) rats were randomly divided into four group( n = 15 ): normal saline group (NC group ), glucocorticoids group (GC group), high-dose protocatechualdehyde group ( GHP group), low-dose protocatechualdehyde group ( GLP group). The rats in NS group were given 0. 9% sodium chloride solution by gavage, the rats in GHP group were given high-dose protocatechualdehyde (8mg·kg^-1·d^-1) by garage, the rats in GLP group were given low-dose protocatechual-dehyde (4mg ·kg^-1·d^-1) by garage, however, the rats in GC group only received prednisone acetate ( 3.5 mg·kg^-1·d^-1). Results The levels of bone physical indexes, bone density,important organ ratio, bone mineral matter and bone organic matter in GC group were significantly decreased, as compared with those in control group ( P 〈 0.05). The levels of bone physical indexes, bone density, important organ ratio, bone mineral matter and bone organic matter in GC group were significantly increased after treatment by high-dose and low-dose protocatechualdehyde, as compared with those in GC group ( P 〈 0.05 ), but there were no significant differences in these parameters between the two treatment groups and NC group (P 〉 0. 05 ). Conclusion Protocatechualdehyde can significantly improve the bone physical indexes, increase bone density, bone mineral matter and hone organic matter, and enhance the important organ ratio, as a result, which can effectively prevent the occurrence of osteoporosis induced by prednisone acetate.
