鸽致病性大肠杆菌的分离鉴定及耐药基因的检测

摘　　要：江苏省某鸽场鸽群出现呼吸困难,死亡增多现象,剖检病死鸽,呈现气囊炎、腹膜炎和皮下广泛性出血等病变。无菌采集脾脏、肺脏等脏器进行病毒分离,但未分离到病毒。选取健康鸽和病鸽各10只,测定新城疫抗体,其抗体水平均较高。同时,无菌采集病料分离到2株细菌,经分离培养、染色镜检、生化特性鉴定,确定为大肠杆菌,玻板凝集试验表明C株为O163型,D株为O138型。对分离到的细菌做药敏试验,结果表明,C株对链霉素等8种药物产生了耐药性,对氟苯尼考等4种药物敏感;D株对庆大霉素等5种药物产生耐药性,对链霉素等7种药物敏感。分别设计引物对其耐药基因进行PCR检测。The pigeons from one farm in Jiangsu province had symptoms of dyspnea and high mortality.The autopsy showed there were air sac inflammation,peritonitis and extensive subcutaneous bleeding.10 sick pigeons and 10 health pigeons were detected respectively for Newcastle disease antibodies and the result demonstrated that the titer were high and there was no difference between the two antibodies.Two strains of bacterium were isolated,cultured,stained,tested by microscopy,biochemical identification.The results showed that the isolates were both Escherichia coli,agglutination test showed that the serotype of the C strain was O163 and the serotype of the D strain was O138.Drug sensitive test showed that C strain was sensitive to 4 drugs such as florfenicol,and was resistance to 8 drugs such as streptomycin.The D strain was sensitive to 7 drugs such as streptomycin,and was resistance to 5 drugs such as gentamicin.Primers were designed and the resistance genes of the two E.coli strains from pigeons were detected.
