摘 要:目的优化青蒿中青蒿素的提取工艺,并为工业化提取青蒿素建立快速测定原料青蒿中青蒿素的含量测定方法。方法使用超声提取法,采用正交设计法优化提取工艺,制得样品溶液;含量测定采用Diamonsil C18柱(250 mm×4.6 mm,5μm),流动相为乙腈-水(体积比68∶32),流速为1.0 mL.min-1,检测波长为210 nm。结果青蒿素在1~40μg范围内与峰面积呈良好的线性关系(r=0.999 9),平均加样回收率为98.59%,RSD值为1.34%(n=6)。结论本提取工艺简便易行;含量测定方法简便、准确、重复性好,适用于青蒿素工业化提取原料的质量控制。Objective To establish an optimal extraction process and a rapid method for the determination of Artemisinin from Artemisia annua L. in industrialization. Methods The sample was treated by ultrasound wave and the orthogonal test was designed to optimize the preparation process. The assay was performed on a C18 column (250 mm×4.6mm,5μm) with a mobile phase consisted of acetonitfile and water (68: 32) , and the flow rate was 1.0 mL ~ min-t~ The detection wavelength was at 210 nm. Results Artemisinin has a good linearity in the range of 1 -40 μg (r =0.999 9) ,and the average recovery was 98.59% with the RSD of 1.34% (n = 6). Conclusion The extraction technology is convenient and feasible. The assay method is simple, accurate and suitable for the quality control of Artemisia annua L. in industrialization.
