摘 要:目的研究大鼠骨髓来源的间充质干细胞(MSCs)对窒息新生大鼠引致的多脏器损伤的影响及相关机制。方法采用贴壁选择法分离、培养、扩增大鼠骨髓来源MSCs,以PBS按要求稀释供注射用;3日龄清洁级SD新生大鼠32只,随机分为窒息+MSCs组(A组)、空气+MSCs(B组)、窒息对照组(C组)、正常对照组(D组)四组;A、C组均先将大鼠置于密封瓶中密闭30min,再予氧罩吸氧2h[氧浓度(60±5)%];B、D组同时置于密封瓶中,但瓶口不密闭,也不予吸氧。模型结束后,A、B组每只大鼠腹腔注射5×104的MSCs,而C、D组仅注射相同容积的PBS,72h后(6日龄)处死全部动物,组织病理学进行心、脑、肺组织病理评分;ELASA法检测心、脑、肺组织匀浆TNF-α含量及血清TNF-α、CK-MB、cTnT、S-100b、NSE水平。结果 (1)心脏、脑、肺组织四组病理学评分分别是1.79±0.22,0.34±0.13,2.33±0.27,0.34±0.08;3.06±0.33,1.29±0.14,4.30±0.47,1.15±0.10;2.31±0.56,0.46±0.20,3.06±0.33,0.44±0.18,差异有统计学意义(P均=0.000)。(2)四组间血清TNF-α、CK-MB、cTnT、S-100b、NSE水平差异存在统计学意义(F=32.19,P=0.000;F=39.14,P=0.000;F=242.40,P=0.000;F=66.66,P=0.000;F=78.66,P=0.000)。(3)以上各指标结果在A、C组均明显高于D组(均P<0.05),C组高于A组(P<0.05)。(4)血清TNF-α水平与细胞因子及酶学改变呈正相关。组织匀浆TNF-α水平与相应组织病理评分也存在正相关(P均<0.05)。结论 (1)新生大鼠窒息模型合并有多脏器损伤。(2)MSC对窒息新生大鼠多脏器损伤具有保护作用,可能与抑制损伤部位的炎症反应有关。Objective To investigate the influence of mouse mesenchymal stem cells on multiple organ damage of new born rats suffered asphyxia.Methods Mouse mesenchymal stem cells were separated,cultured,amplificated.Thirty-two three-day old Sprague-Dawley(SD)rats were randomly divided into four groups according to their exposure condition and with or without administration of mMSCs,group A and group C rats were placed in sealed bottles sealed for 30 min,then exposed with oxygen concentration of(60±5)%;group B and group D also placed in sealed bottles at the same time,but the bottle was not closed.Then they were injected intraperitoneally with MSCs of 1×105 or phosphate buffered solution(PBS)respectively.72 h later,all of them were killed,pathological scores of heart,brain and lung were counted for the histopathological study under light microscope;the levels of TNF-α from heart,brain and lung homogenates and the levels of TNF-α,CK-MB,cTnT,S-100b and NSE from serum were detected by ELISA.Results The pathological scores of heart,brain and lung in four groups were 1.79±0.22,0.34±0.13,2.33±0.27,0.34±0.08;3.06±0.33,1.29±0.14,4.30±0.47,1.15±0.10;2.31±0.56,0.46±0.20,3.06±0.33,0.44±0.18,respectively.There were significantly differences in the level of TNF-α in the homogenate of hearts,brains and lungs among four groups.All indicators above in group A and group C were significantly more than those in group D(all P0.05),while these indicators in group A were lower than those in group C.There were positive correlation between TNF-α and CK-MB,cTnT,S-100b and NSE in serum respectively.There were also positive correlation between TNF-α and pathological score in different organs(r=0.753,P=0.000;r=0.842,P=0.000;r=0.956,P=0.000).Conclusions The model of neonatal rats with asphyxia result in the damage of heart,brain and lung;MSCs injected intraperitoneally have effective protection for mutlti-organ damage suffered asphyxia.
