摘  要:目的研究阿托伐他汀对心肌缺血复合冷应激大鼠左室心肌组织中Bim和Caspase-3表达的影响。方法 42只雄性SD大鼠随机分为:心肌缺血组、心肌缺血复合冷应激组、阿托伐他汀组。以永久性左冠状动脉前降支结扎术+4℃冷刺激(8 h/d,4 d)建立心肌缺血复合冷应激模型。阿托伐他汀组复合应激前3 d及后4 d给以阿托伐他汀灌胃(20 mg.kg-1.d-1)。实验结束时观察血脂水平,超声心动图评价左室收缩功能,TTC染色法测定心肌梗死面积,Western印迹法检测心肌组织Bim、Caspase-3蛋白表达。结果各组大鼠血脂指标无统计学差异。心肌缺血复合冷应激使梗死面积增大(P<0.01)、左室收缩功能降低;阿托伐他汀干预后心功能改善、梗死面积缩小(P<0.01),Bim、Caspase-3表达下降(P<0.01)。结论阿托伐他汀改善心肌缺血复合冷应激大鼠左室收缩功能,其机制可能通过下调Bim、Caspase-3表达实现。Objective To investigate the effect of atorvastatin on the expression of Bim and Caspase-3 in left ventricular myocardial tissue of rats following co-stress of myocardial ischemia and cold. Methods 42 male SD rats were divided into ( n = 14) : ischemia, ischemia + cold stress and atorvastatin groups. Co-stress model was induced by ligation of left coronary artery combined with cold stress of4℃, 8 h/ d for 4 consecutive days. Rats were gavaged with atorvastatin 20 mg· kg^-1 · d^-1 on 3 days pre- co-stress and 4 days post-co-stress. At the end of the experiment, levels of serum lipids were measured, left ventricular systolic function was assessed by echocardiography ; myocardial infarct size was determined by TTC staining; Bim and Caspase-3 expression in myocardium was determined by Western blotting. Results There was no significant change in blood lipids in each group. Co-stress of myocardial ischemia and cold stress significantly deteriorated the cardiac function and increased the infarct size (P 〈 0.01 ) , which was attenuated by atorvastatin by decreasing Bim, Caspase-3 expression ( P 〈 0. 01 ). Conclusions Atorvastatin could improve left ventricular systolic function of rats following co-stress of myocardial ischemia and cold, one of the mechanisms of the effect is related to downregulating the overexpression of Bim and Caspase-3 in left ventricular myocardial
