摘 要:目的 观察氧因素对趋磁螺菌AMB-1生长量、磁小体形成及其相关基因表达的影响,为确定磁小体合成最适条件和研究磁小体形成机制提供实验依据.方法 在地磁有铁条件下,AMB-1经培养驯化接种,分别置于好氧和微好氧条件下培养.定时取样检测菌液OD600和Cmag值来研究细胞生长量和磁性增长量;用透射电镜观察16h菌样的磁小体形态;qRT-PCR检测磁小体相关基因mms13,magA和mms6的表达.结果 有铁微氧种子转至好氧条件培养12h后,其OD600值较对照组显著升高,2.5h后Cmag值明显降低,磁小体链短而疏松.16h菌样mms13,magA,mms6基因表达无明显差异.有铁好氧种子转至微氧条件培养14h后,其OD600值较对照组显著降低,10h后Cmag值显著增高,磁小体呈紧密的长链分布,16h菌样mms13基因表达量增加6倍,magA和mms6明显增高.结论 好氧条件有利于AMB-1细胞的生长,微氧条件促进磁小体的形成,并明显影响mms13,magA和mms6基因表达.Objective To invesligale effects of oxygenlension on magnetosome formation and expression of nmlsl3,mms6 and magA in Magenetospirilhtm magneticum AMB-I. 11 can provide some basis fl^r determing the optimum magnetosome synthesis conditions and studing magnetosome fin'marion mechanism. Methods Under geomagnetic iron condition,the trained AMB-I were inoculated Io EMS(,M and cuhivated in micro-aerobic and aerobic ~,onditions, respecliwdy. To research the effects of oxygentension on growth and magneh^some forma- tion,regularly lest ()l)t,~~ and C,,,,,~ values, and ohserve the ~'ell witll transmission electron microscope. The expression of mm^d3, mm^6 and magA which were related to magnelosome formation by qRT-PCR were analyzed. Results In aerobiccondition,OI)e^x~ increased rapidly after twelve hours and C,,,,,~ value had a obvious drop after 2.5 hours,at 161h the expression of mms l3 ,m.gA and rams6 had no signifi~.ant difl^r- ence. Magneloso,ne chain was short and loose. In micro-aerobic condition,OD600 increased significantly from Ihe 14lh hour and C xalue re- duced obviously after 10 hours,at the 16/h hour the expression of mms l3 increased 6 times,magA and mm.d6 increased signifintly. Magnetosome shaped in loselv long-chain. Conclusion Aerobic condition can facilitate the cell growth,micro-aerobic promote maguetosome firmation and effect the expression of mms l3,magA and mm.~6 in Magenet.spirilhml magnetivum A MB -1.
