摘 要:目的通过观察中药复方强心复脉颗粒对损伤兔窦房结组织缝隙连接蛋白45(Cx45)表达的影响,探讨其治疗病态窦房结综合征的可能机制。方法 30只大耳白兔随机分为正常组,模型组,阿托品组,强心复脉颗粒高、中、低剂量组(以下简称高、中、低剂量组),每组5只。除正常组外,余各组均采用甲醛加压注射渗透法建立兔窦房结组织损伤模型,待模型稳定后各用药组灌胃给药7 d,免疫组织化学方法检测各组兔窦房结组织Cx45表达的变化。结果与正常组相比,模型组兔窦房结组织Cx45表达量显著降低(P<0.01);与模型组相比,高剂量组家兔窦房结组织Cx45表达量显著升高,且明显优于中、低剂量组及阿托品组(P<0.05或P<0.01);阿托品及中、低剂量组之间无明显差异(P>0.05)。结论窦房结组织损伤后Cx45表达量明显减少,强心复脉颗粒能上调损伤窦房结组织Cx45的表达,可能是其治疗病态窦房结综合征的分子机制之一。Objective To study the effects of the Chinese medicine Qiangxin Fumai Granule on the expression of Connexin45(Cx45) of sinoatrial node(SAN) cell damages in rabbits,and to discuss the mechanism of Qiangxin Fumai Granule in treating sick sinus syndrome.Methods Thirty big ears white rabbits were randomly divided into 6 groups:the normal group,the model group,the atropine group,the high-dose Qiangxin Fumai Granule group,the middle-dose Qiangxin Fumai Granule group and the low-dose Qiangxin Fumai Granule group,with five animals in each group.The model was established by injecting formaldehyde to SAN except the normal group.After the model established,the rabbits received intragastric administration for 7 days.The expression of Cx45 in SAN was determined by immunohistochemistry.Results Compared with the normal group,the expression of Cx45 in SAN of model group reduced markedly(P0.01).Compared with the model group,the expression of Cx45 increased markedly in high-dose Qiangxin Fumai Granule group,which was superior to the middle-dose Qiangxin Fumai Granule group,low-dose Qiangxin Fumai Granule group and atropine group(P0.05 or P0.01).There was no significant difference among the middle-dose Qiangxin Fumai Granule group,low-dose Qiangxin Fumai Granule group and atropine group(P0.05).Conclusion The sinoatrial node tissue injury significantly reduced Cx45 expression,and Qiangxin Fumai Granule can improve the expression of Cx45 in SAN,which may be related to the treatment of sick sinus syndrome.
