摘  要:目的:研究MYH9和ABI2基因的单核苷酸多态性(SNP)在2型糖尿病人群中与血糖、胰岛素抵抗及高血压的关系。方法:应用基质辅助激光解吸电离飞行时间质谱技术,对773例天津地区的2型糖尿病患者的MYH9基因的8个SNP位点和ABI2基因的4个SNP位点进行基因分型,以PLINK软件进行数量性状的关联分析及基因型交互作用分析。结果:MYH9和ABI2基因SNP位点与血糖和胰岛素抵抗表型存在较广泛的关联,包括糖化血红蛋白、2 h血糖、空腹和2 h胰岛素及胰岛素抵抗指数(P<0.05)。ABI2基因SNP位点rs62183937(P=0.000),rs11675251(P=0.008)等均与血压相关。结论:MYH9和ABI2基因的多态性可能与2型糖尿病患者的血糖、胰岛素抵抗相关表型及2型糖尿病合并高血压的发病相关。Objective: To investigate the relationship of MYH9,ABI2 gene single nucleotide polymorphisms(SNPs) and glucose,insulin resistance,and hypertension in type 2 diabetes patients.Methods: Eight SNPs in MYH9 gene and 4 SNPs in ABI2 gene were genotyped in 773 type 2 diabetes patients in Tianjin area by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDITOF MS).Plink was used for quantitative association studies and genotype interactions(epistasis) analyses.Results: Significant associations were found among MYH9 and ABI2 SNPs and insulin resistance phenotypes,including HbA1c,2-hr glucose,fasting and 2 hr plasma insulin(P0.05).Moreover,association was found among ABI2/MYH9 SNPs and affection status of hypertension(rs62183937 and rs11675251 in ABI2,P=0.000,P=0.008,respectively).Conclusion: It is suggested that polymorphisms in these 2 genes are associated with glucose,insulin resistance,and hypertension related phenotypes in type 2 diabetes patients.
