摘 要:目的研究枳术通便汤对慢传输型便秘(slow transit constipation,STC)大鼠肠神经系统、结肠神经营养因子(glial cell line derived neurotrophic factor,GDNF)及一氧化氮合酶(nitric oxide synthase,NOS)mRNA表达的影响。方法建立STC大鼠模型30只,随机分为模型组及中药高、低剂量组,每组10只,另选取10只大鼠作为空白对照组(空白组)。中药高、低剂量组分别予枳术通便汤[生药4.8g/(kg·d)、2.4g/(kg·d)]灌胃,空白组及模型组给予等量生理盐水灌胃。采用墨汁推进试验测定大鼠肠道墨汁推进率,采用RT-PCR法测定结肠GDNF及NOS mRNA表达,比较各组结果。结果与空白组比较,模型组墨汁推进率及GDNF mRNA表达降低,NOS mRNA表达升高,差异有统计学意义(P<0.01,P<0.05);与模型组比较,两个中药组墨汁推进率升高,中药高剂量组GDNF mRNA表达升高,NOS mRNA表达降低,差异有统计学意义(P<0.01,P<0.05);中药两剂量组各指标比较,差异无统计学意义(P>0.05)。结论高剂量枳术通便汤对STC大鼠肠道传输有明显改善作用,其作用机制可能是通过提高结肠GDNF mRNA表达,降低NOS mRNA的表达而促进肠道的动力。Objective To observe the effects of Zhizhu Tongbian Decoction(ZTD) on the enteric nervous system,mRNA expressions of glial cell line derived neurotrophic factor(GDNF) and nitric oxide synthase(NOS) in the slow transit constipation(STC) rats.Methods Thirty STC rat model was established by gastric irrigation of rhubarb.After the model building,they were randomly divided into three groups,i.e.,the model group,the high dose ZTD group,and the low dose ZTD group,10 in each.Another 10 rats were selected as the blank control group.Rats in the high dose ZTD group and the low dose ZTD group were administered with ZTD(at the daily dose of crude drug 4.8 g/kg and 2.4 g/kg respectively) by gastrogavage.Normal saline was given to rats in the blank control group and the model group.The ink propelling rate was determined using ink propelling test.Meantime,mRNA expressions of GDNF and NOS in the rat colon were measured using reverse transcriptional polymerase chain reaction(RT-PCR).Results Compared with the blank control group,the ink propelling rate and GDNF mRNA expression decreased,and NOS mRNA increased in the model group,showing statistical difference(P0.01,P0.05).Compared with the model group,the ink propelling rate increased in the high and low dose ZTD groups(P0.01,P0.05).The mRNA expressions of GDNF increased and the mRNA expressions of NOS decreased in the high dose ZTD group with statistical difference(P0.01,P0.05).But there was no difference in any index between the high and low dose ZTD groups.Conclusion High dose ZTD could obviously improve the intestinal transmission function possibly through up-regulating the mRNA expressions of GDNF and down-regulating the mRNA expressions of NOS in STC rats.
