中国汉族人群PPARγ2基因Pro12Ala多态性与2型糖尿病关联的Meta分析

摘　　要：目的探讨中国汉族人群过氧化物酶体增殖物激活受体y2（peroxlsome proliferator—activated receptor—y2，PPARy2）基因Pro12Ala多态性与2型糖尿病（type2diabetes mellitus，T2DM）易感性的关联。方法系统检索PubMed、中国生物医学文献数据库、中国期刊全文数据库及万方数据库文献，检索时间范围为1990年1月至2011年6月，收集关于中国汉族人群PPARy／2基因Pro12Ala多态性与T2DM易感性关联研究的所有文献，共检索到29篇文献，对各研究结果进行异质性检验和效应值合并，同时进行敏感性分析。结果共纳入21篇文献，累计T2DM患者3870例，对照3333名。总体研究人群中T2DM组和对照组合并的Ala等位基因频率分别为4．13％（320／7740）、4．56％（304／6666）；纳入的各研究结果间同质性较好（x2=25．96，P=0．17）。T2DM组与对照组（PA＋AA）／PP基因型频率合并OR（95％CI）值为0．96（0．81～1．14）（P=0．64）。剔除最大的权重研究文献及较大Ala等位基因频率研究文献后，同质性较好（x2值分别为24．23、16．87，P值均〉0．05）；OR（95％CI）值分别为1．01（0．84～1．21）和1．07（0．89～1．28），P值均〉0．05。结论PPARy2基因Pro12Ala多态性与我国汉族人群T2DM易感性无关联。Objective To evaluate the relationship between peroxisome proliferator-activated receptur-y2 （PPARy2） Pro12Ala polymorphism and type 2 diabetes mellitus （T2DM） in Chinese Han population. Methods PubMed, Chinese Biomedicine Database （CBM）, China National Knowledge Infrastructure （CNKI） and Wanfang database were searched for all relevant articles investigating the association between PPAR3,2 Pro12Ala polymorphism and T2DM that were available fi＇om .January, 1990 to June, 2011. A total of 29 relevant articles were selected. A Meta-analysis was performed to estimate heterogeneity and the pooled odds ratio （OR） to evaluate the relationship between PPARy2 Pro12Ala polymorphism and T2DM. The sensitivity analysis was also assessed. Results A total of 21 qualified articles including 3870 patients with T2DM and 3333 healthy controls were analyzed in the study. The frequencies of the allele Alal2 in T2DM and control groups were 4. 13% （320/7740） and 4. 56% （304/6666）, respectively. There were not heterogeneity （ x22 = 25.96, P = 0. 17 ） among the 21 qualified articles. The pooled OR （95% CI） value of the frequencies of the PPARy2 genotype （PA ＋ AA）/PP calculated by fixed effects model was 0. 96 （0. 81 - 1.14） （ P = 0. 64）. There was not heterogeneity among the remaining articles after excluding the article with the largest weight and the article with larger frequencies of the allele Alal2 respectively （ X2 values were 24. 23, 16.87 respectively, both P values 〉 0. 05）. The pooled OR （95 % CI） value of the frequencies of the PPARy2 genotype （ PA ＋ AA ）/PP of the remaining articles were 1.01 （0. 84 - 1.21 ） and 1.07 （0. 89 - 1.28） after excluding the article with the largest weight and the article with larger frequencies of the allele Alai2 （both P values 〉 0. 05）. Conclusion PPARy/2 Pro12Ala polymorphism was not associated with type 2 diabetes mellitus in Chinese Han population.
