摘  要:目的:通过观察细胞因子指标,研究丹参酮及电针对脊髓缺血再灌注损伤免疫途径的干预作用。方法:将新西兰家兔24只,随机分为模型组、丹参酮组、电针组和丹参酮+电针组。采用Zivin法改进复制模型,于造模前1h给予相应治疗。各组家兔分别于治疗前、缺血30min后再灌注0.5、1、4、8、12h分别采家兔股静脉血。检测血清白介素1β(IL-1β)、白介素1受体(IL-1Ra)、白介素8(IL-8)指标。结果:与造模前比较,模型组家兔血清IL-1β、IL-1Ra、IL-8再灌注各时点均升高(P<0.01)。3种治疗手段在不同时间点抑制IL-1β、IL-8的升高(P<0.05);促进IL-1Ra的升高(P<0.05);在再灌注1h点,丹参酮+电针组抑制IL-8的升高效果优于丹参酮组及电针组(P<0.05)。结论:家兔脊髓缺血再灌注损伤后,可导致家兔血清IL-1β、IL-1Ra、IL-8升高。丹参酮及电针对脊髓缺血再灌注损伤免疫途径的干预作用是通过抑制IL-1β、IL-8、促进IL-1Ra的升高,而起到治疗作用。丹参酮+电针治疗手段疗效优于单用丹参酮或电针,针药结合的优势明显。3种治疗手段存在时间窗效应。Objective: To investigate the effect of tanshinone and/or electro-acupuncture on IL-1β,IL-1Ra and IL-8 in serum of rabit with spinal cord ischemia reperfusion injury.Methods: Twenty-four New Zealand rabbits were randomly divided into ischemia-reperfusion group,Tanshinone intervention group,electroacupuncture group and tanshinone plus electroacupuncture group.Blood was collected before replicating model and 0.5h,1h,4h,8h,12h after reperfusion by rabbit femoral venous,based on which IL-1β,IL-1Ra and IL-8 levels were examined.Results: In ischemia-reperfusion group,significant increase of IL-1β,IL-1Ra and IL-8 levels were observed at each time point compared with the baseline level(P0.01).TanIIa and/or electro-acupuncture can inhibit the elevation of IL-1 and IL-8(P0.05).In addition,TanIIa and/or electro-acupuncture can promote the increase of IL-1RA(P0.05) at 4h after reperfusion(P0.05).Significant decrease of IL-8 was observed in TanIIa plus electro-acupuncture at 1h after reperfusion and 12h after reperfusion,respectively(P0.05).Conclusion: In model group IL-1β,IL-1RA,IL-8 levels were higher than the levels before replicating model.Tanshinone and/or electroacupuncture played an important role in the intervention of the immune pathway of spinal cord ischemia-reperfusion injury by inhibiting the levels of IL-1 and IL-8 together with promoting the elevation of IL-1RA.Tanshinone plus electroacupuncture treatment were more efficient compared with the other two interventions.Moreover,time window effect was observed in TanIIa and/or electro-acupuncture.
