基于均匀设计与灰色关联分析的中药碧血胶囊成分配伍组方研究

摘　　要：目的:优化中药碧血胶囊成分配伍方,探讨其对心肌细胞生存率和细胞凋亡的影响。方法:将中药碧血胶囊的4个成分按四因素六水平的均匀设计,对异丙肾上腺素诱导大鼠心肌细胞损伤模型进行了实验干预。采用MTT法检测细胞生存率,流式细胞仪和细胞荧光染色检测心肌细胞凋亡,并通过灰色关联分析其成分配伍方的主次关系。结果:均匀设计得出优化的成分配伍方100μmol/L人参皂苷Rb1+80μmol/L绞股蓝皂苷Ⅳ+15μmol/L荭草苷。灰色关联分析获得该方中各成分的效应关系为绞股蓝皂苷Ⅳ>荭草苷>人参皂苷Rb1。优化的成分配伍方提高细胞生存率作用明显优于单一成分组,其抑制心肌细胞凋亡的效应优于碧血胶囊药物血清(P<0.01)。结论:通过均匀设计与灰色关联分析以及实验验证获得了中药碧血胶囊的优化成分配伍方及其成分效应的主次关系。Objective： To optimize the combination of Bixue Capsule components,and to discuss its effects on the myocardial cell survival rate and apoptosis.Methods： Four components of Bixue capsule according to the 4 factors and 6 level of uniform design were experimentally treated to isoproterenol（ISO） induced rats myocardial cell injury model.The cell survival rate was detected by MTT assay.The myocardial cell apoptosis were measured by fluorescence staining and flow cytometry assays,and the primary and secondary relations of components-combined formula were testing by the grey relation analysis.Results： The uniform design optimized formula was 100μmol/L Ginsenoside Rb1＋80μmol/L Gypenosides Ⅳ＋15μmol/L Orientin.The grey correlation analysis showed that effective relation in the formula was Gypenosides Ⅳ﹥Orientin﹥Ginsenoside Rb1.The improved cell survival rate by the optimized components-combined formula was better than each component alone,and the effect on the inhibition of myocardial cell apoptosis was better than Bixue capsule serum（P0.01）.Conclusion： Through the uniform design,grey relational analysis and experimental confirmation,it was demonstrated that the optimization of Bixue Capsule components-combined formula,and the primary and secondary relations of effect on each components of the formula.
