摘 要:应用PHA、ConA刺激经IFN-α,rIFN-γ作用24小时后的正常人外周血单个核细胞,观察其对产生IL-2、IF N-γ能力与环核苷酸含量的影响。结果表明rFN-α、IFN-γ对单个核细胞产生IL-2、IFN-γ能力的影响取决于外源性IFNs的剂量,随作用的IFN-α、rIFN-γ剂量增加,渐出现抑制性效应。IFNs作用后的单个核细胞诱生IL-2、IFN-γ能力之间有较好的相关关系,提示它们关系密切。此外,IFN-α,rIFN-γ能够增加单个核细胞内cAMP含量与cAMP/cGMP比值,提示这一作用可能同IL-2、IFN-γ等免疫调节因子的细胞生物学调节效应有关。When the peripheral blood lymphocytes treated by IFN-α and rIFN γ for 24 hrs from the healthy donors were stimulated by PHA and Con A. the abilities of producing IL-2 and IFN-γ and the levels of the cyclic nucleotides were studied in the treated lymphocytes. The results showed that the effects of IFN-α and rIFN-γ on the abilities of producing IL-2 and IF N-γ were dose-dependent, and as the doses of IFN-αand rIFN-γ were increased the inhibiting action was found eventually. It was suggested that the abilities of producing IL-2 and IFN-γ in the IFNs-treated lymp hocytes be two events correspondingly. In abbition, IFN-α and rIFN-γ could increase intracellular levels of cAMP and cGMP as well as the ratio of cAMP to cGMP, which revealed that the results mentioned above should relate to the action of IL-2, IFN-γ and other lymphokines which possessed the biological function in immune modulation.
