摘 要:由单核-巨噬细胞产生的白细胞介素1(Interleukinl,IL—1)在免疫应答及炎症反应的分子调节网络中起着重要作用,与许多临床疾病的发病有关。A variety of stimulants including immune complex and aggregate immunoglobu(?)ins (AIgs) have the potential to stimulate Mφ producing IL-1.we explored whethe r or not AIgA and/or AIgG stimulate IL-1 production. Heat-aggregated human IgA (MW 600 to 1000KD) was prepared under sterile conditions. Resident rat peritoneal MφS were obtained by peritoneal lavage.AIgA, IgA, AIgG, IgG were added into Mφ suspension respectively. IL-1 like activity was determined by ~3H-thymidine incorporation in SD rat thymocytes. IL-2 actiyity was excluded by assay with IL-2-dependent cells (?). It was shown in 9 samples from each group that only AIga had the effect on stimulating Mφ to produce IL-1 like factor [CPM mean±SD: (1) AIgA 4,845±2, 135: (2) AIgG 2 ,145±491; (3) IgA 1.447±30b (4) IgG 1, 602±290; (5) No stimulants 1,463±158; (1) vs (2)-(5) P<0.00l] . The effect of A-IgA was comparable to that of Lipopolysaccharide (LPS) 3,162±351. This outcome is likely due to the present of receptors which highly bind with poly-IgA. This result has therapeutic implications for the pathogenetic effect of poly-IgA .
