摘  要:目的:通过研究对胸腺细胞发育具有正性调节的作用的转录因子TCF-1和负性调节作用的Nur77在6周递增负荷运动的应答性特征,探讨长期运动影响胸腺细胞发育的调节机制。方法:128只SD大鼠随机分为运动组和对照组,运动组进行递增负荷跑台训练6周,每周6次,每天30 min。分别于第02、、46、周末利用IHC、WB及FQ-RT-PCR技术测定安静状态、运动后即刻和运动后3 h胸腺TCF-1、Nur77及其mR-NA表达水平。结果:TCF-1和Nur77呈现几乎相反的应答性变化:1)TCF-1:对负荷初次应答时运动前后没有明显变化;第2周末和第四周末运动后明显降低,恢复3 h升高,呈"V"型应答曲线;第6周末,则在运动后持续下降。其mRNA应答趋势与其蛋白表达相一致。2)Nur77:第0、2、4周末呈现倒"V"型变化,第6周则持续上升。其mRNA应答趋势与其蛋白表达相一致。结论:随着递增负荷运动的进程,TCF-1及其mR-NA总体水平越来越低,且越来越难以恢复,表明对胸腺细胞发育的正性调节作用越来越弱;Nur77及其mR-NA总体水平越来越高,恢复也越来越难,表明对胸腺细胞的负性调节作用越来越强。这应该是胸腺细胞受长期递增负荷运动影响,胸腺细胞发育受阻且功能逐步下降的重要调节机理之一。objective:This study tried to observe responsive characteristics of the positive regulatory factor,TCF-1,and negative factor,Nur77,to thymus lymphocyte development during six weeks incremental exercise so as to shed some light on regulative mechanism of thymocyte development with long-term exercise.Method: 128 rats,were randomly divided into control and exercise group.Exercise protocol: the exercise group conducted load incremental step running exercise for six weeks,30 minute a day and six times a week.Samples were taken before exercise and in the end of WK2,4 and 6,at rest,just after exercise and 3 hours after exercise,respectively.TCF-1,Nur77 and their mRNA in thymus were measured by IHC,WB and FQ-RT-PCR.Result: TCF-1 and Nur77 presented almost oppositely response during six-week incremental exercise: 1) TCF-1: no distinguished changes occurred in pre-and post-exercise,and "V" responsive pattern were observed in week 2 and 4.And its mRNA showed similar changing pattern.2) Nur77: response showed an inversed "V" pattern in the end of WK0,WK2 and WK4.Conclusion: With incremental exercise,level of TCF-1 and mRNA were gradually decreased,and showed more difficult recovery while Nur77 and mRNA showed opposite change.This indicated down-regulation to thymus lymphocyte development was stronger and stronger with exercise,on the contrast,up-regulation weaker and weaker.As result of interaction of two factors,lymphocyte's development was seriously inhibited.This should be one of the important mechanisms of exercise-induced immunosuppression.
