摘 要:目的观察丹参酮ⅡA对血管紧张素Ⅱ(AngⅡ)诱导的心肌成纤维细胞Ⅰ型、Ⅲ型胶原合成的影响及可能的机制下调CTGF水平。方法采用AngⅡ诱导和建立心肌成纤维细胞纤维化模型,采用不同浓度的丹参酮ⅡA处理纤维化细胞,观察其对Ⅰ型、Ⅲ型胶原mRNA的影响,同时采用Western blot和RT-PCR检测CTGF和CTGF mRNA水平。结果 (1)AngⅡ处理后的心肌成纤维细胞的Ⅰ型、Ⅲ型胶原的mRNA水平高于DMSO组,且给予丹参酮ⅡA处理后,Ⅰ型、Ⅲ型胶原mRNA水平呈浓度依赖的方式下降,且在50μmol/L的浓度下,恢复正常;(2)AngⅡ处理后的心肌成纤维细胞的CTGF和CTGF-mRNA水平高于DMSO组,且50μmol/L丹参酮ⅡA可降低CTGF和CTGF-mRNA水平;(3)给予外源性的CTGF处理后,丹参酮ⅡA抑制成纤维细胞胶原合成的作用减弱,且10μmol/L的CTGF可完全减弱丹参酮ⅡA的作用,外源性CTGF处理未改变CTGF-mRNA水平。结论丹参酮ⅡA具有降低血管紧张素Ⅱ(AngⅡ)诱导的心肌成纤维细胞Ⅰ型、Ⅲ型胶原合成的作用,可能的机制是下调CTGF水平。Objective: To explore the influence of Tanshinone Ⅱ A on the type I and Ⅱ collagen synthesis of CFB induced by angiotensin Ⅱ (Ang Ⅱ ) as well as the possible mechanism of down-regulating CTGF level. Methods:The CFB fibrosis model was induced by angiotensin II stimulation and treated by different concentra- tions of Tanshinone Ⅱ A. The following items were investigated such as type Ⅰ , Ⅲ collagen mRNA levels. Mean- while, Western blot and RT-PCR were employed to detect CTGF and CTGF-mRNA separately. Results:The type I and the Ⅲ collagen mRNA levels of CFB stimulated by Ang Ⅱ were higher than those of the DMSO group, while the tanshinone Ⅱ A treatment can decrease the I and the Ⅱ collagen mRNA levelin a concentration-depen- dent manner. The collagen mRNA level return to normal at 50μmol/L tanshinone Ⅱ A. The CTGF and the CT- GF-mRNA levels of CFB stimulated by Ang Ⅱ were higher than those of the DMSO group. The tanshinone Ⅱ A can reduce the level of CTGF and CTGF-mRNA. After the administration of exogenous CTGF, the anti-collagen synthesis effects of tanshinone Ⅱ A were attenuated with the maximum effect at 10μmol/L,while exogenous CTGF treatment did not change CTGF-mRNA levels. Conclusion:Tanshinone Ⅱ A can lower the type I and Ⅲ collagen synthesis of cardica fibroblasts induced by Ang Ⅱ with possible mechanism by reducing CTGF levels.
