摘  要:以苜蓿雄性不育株系及其可育株系的花药为外植体,研究取材时间和不同激素配比对其愈伤组织诱导及分化的影响。结果表明,雄性不育株系花药培养时,以现蕾初期取材、MS为基本培养基的条件下出愈率较理想。愈伤组织诱导的适宜培养基为:MS+0.5mg/L 2,4-D+0.25mg/L 6-BA+0.2mg/L NAA+3mg/L KT+3%蔗糖+0.7%琼脂(出愈率66.7%);不育与可育植株花药愈伤适宜的分化培养基分别为:MS+1mg/L KT+0.2mg/L NAA+2%蔗糖+0.7%琼脂(分化率75%)和MS+4mg/L KT+0.2mg/L NAA+2%蔗糖+0.7%琼脂(分化率79%)。The anther of the male sterile and fertile lines of alfalfa was used for explants in order to study the effect of sampling time and hormone ratio on callus induction and differentiation.The results showed that anther collected in bud stage and induced on the MS medium could get a higher callus induction rate.MS + 0.5mg/L 2,4-D + 0.25mg/L 6-BA + 0.2mg/L NAA + 3mg/L KT +3% sucrose + 0.7% ager was suitable for callus induction(induction rate: 66.7%).MS + 1mg/L KT + 0.2mg/L NAA + 2% sucrose +0.7% ager(differentiation rate: 75%) and MS + 4mg/L KT + 0.2mg/L NAA + 2%sucrose + 0.7% ager(differentiation rate:79%) was suitable for the male sterile and fertile callus differentiation.
