摘  要:为探索适合产业化的仙人掌多糖(Opuntia dillenii Haw.polysaccharides,ODPs)提取工艺,采用单因素及正交试验优化酸法提取ODPs条件;优化的苯酚硫酸法测定ODPs得率及含量;并用酶法结合盐酸法除蛋白,经透析,洗涤得到精制的ODPs。得到酸法提取ODPs最佳条件为:加入浓度为0.10 mol/L的硫酸溶液,液料比为20∶1(mL/g),70℃提取2次,每次2 h。平均产率达39.92%,多糖纯度达87.11%,多糖中蛋白含量降低至1.06%。To explore an extraction method which is suitable to the industrialization,the extraction conditions of ODPs were optimized by the single-factor test and the orthogonal test,and the yield rate and content of ODPs were determined by the phenol-sulfuric acid method optimized by us.The refined ODPs were obtained after removing proteins by enzymatic method and hydrochloric acid method,dialyzing and washing by organic solvents.Results showed that optimum conditions of ODPs extracted by acid method were: the concentration of sulfuric acid 0.10 mol/L,ratio of liquid to material 20∶1(mL/g),extraction temperature 70 ℃,and extraction time 2 h for 2 times.Under these conditions,the mean yield and purity of ODPs were 39.92 % and 87.11 % respectively,and the protein content of ODPs decreased to 1.06 %.
