摘 要:为了探明生物反应器内大花蕙兰原球茎增殖生长的影响因素,为大花蕙兰种苗生产中大量培养繁殖材料提供理论依据,以大花蕙兰茎尖诱导的原球茎为外植体,研究了接种量、蔗糖浓度、活性碳浓度对5 L气升式反应器内原球茎增殖和生长的影响。结果表明,大花蕙兰原球茎在5 L反应器内增殖时,接种量为20 g的处理原球茎鲜重显著优于10 g和30 g接种量处理;添加蔗糖30 g/L处理对大花蕙兰增殖生长有促进作用;活性炭浓度0.25 g/L处理原球茎的生长良好,高于0.25 g/L抑制原球茎的生长,增殖系数为7.5。利用5 L气升式生物反应器培养原球茎,接种量为20 g、蔗糖浓度为30 g/L、活性炭浓度为0.25 g/L时,大花蕙兰原球茎增殖较快,生长较好。In order to explore the influencing factors on proliferation growth of Cymbidium hybridum protocorm by bioreactor culture, which provided theoretical basis for large scale production of C. grandiflorium, the protocorm induced by C. hybridum shoot tip as explants, the effects of inoculation density, sucrose concentration and activated carbon concentration on PLBs proliferation and growth of protocorm were studied in 5 L air-lift bioreactor. The results showed that the protocorm fresh weight of inoculation of 20 g was better than that of 10 g and 30 g in 5 L bioreactor for PLBs proliferation, 30 g/L sucrose was helpful for PLBs proliferation, and the optimal concentration of active carbon (AC) for PLBs growth was 0.25 g/L and higher concentration of AC restricted the growth of PLBs, its multiplication coefficient was 7.5. The growth and proliferation of protocorm were better with inoculation 20 g, sucrose 30 g/L and AC 0,25 g/L during 5 L air-lift bioreactor culture
