虎舌红组织培养中有效无菌材料的获得研究

摘　　要：[目的]探索获得虎舌红(Ardisia mamillata Hance)组织培养中有效无菌材料的途径和方法。[方法]以虎舌红为试材,研究了外植体母本的预处理和外植体的采集、灭菌、接种等处理方式对获得有效无菌材料的影响。[结果]2月将母株移入室内,进行预处理,3月中旬取当年生枝条和萌蘖条上的茎尖,用去污剂(肥皂水或洗衣粉水)浸泡并刷洗后,流水冲洗1～2 h,用75%乙醇表面消毒30 s,无菌水冲洗2～3次后放入0.1%升汞中灭菌(先用0.1%升汞消毒4 min,无菌水冲洗3次,再用0.1%升汞消毒4 min,无菌水冲洗5～8次)后进行有效接种,这种方法能有效降低污染率,提高成活率。[结论]该研究可为进一步研究和大规模生产虎舌红提供技术支持。[Objective] The purpose was to explore the way of obtaining effective non-bacteria material of Ardisia mamillata Hance among tissue culture.[Method] The effects of pretreating of explant female parent and explant collecting,sterilizing and inoculating etc on obtaining effective non-bacteria material of Ardisia mamillata Hance.[Result] The female plant is pretreated in the door in February,then stem tips of current year＇s branches and sprouting branches are collected in mid-March and they are cleaned with eradicator,then are washed by the tap water for one or two times.Explants are sterilized in 70% alcohol for 30 seconds,and are sterilized in 0.1% mercuric chloride（firstly sterilized in 0.1% mercuric chloride for 4 minutes,secondly sterilized in 0.1% mercuric chloride for 4 minutes after washed by the tap water for three times,finally washed by the tap water for 5-8 times） after washed 2-3 times by using sterile water.This way can effectively decreased pollution rate and increased survival rate.[Conclusion] The study provides a technological support for further researching and producing in a large scale of Ardisia mamillata Hance.
