摘  要:[目的]研制Ⅰ型胶原(collagen type I,CI)海绵/血管内皮生长因子(vascular endothelial growth fac-tor,VEGF)缓释生物支架材料,了解该种材料中的VEGF体外缓释效果。为骨肌腱组织工程寻求一种新型复合支架材料。[方法]从CI海绵自身的力学、孔径、紫外最大吸收光谱、孔隙率、含水率、傅立叶红外光谱及与VEGF的组织相容性等方面研究VEGF胶原缓释系统支架材料的特性,利用生物学方法将VEGF负载于CI,制备具有VEGF缓释效果的复合支架。[结果]CI海绵能够成功的将VEGF包裹起来,可以达到明显的缓释作用,与VEGF有良好的组织相容性。[结论]CI海绵有理想的孔径和合理的生物力学及与VEGF良好的生物学相容性结构性质,VEGF可以从CI-VEGF缓释材料中缓慢释放,有明显的缓释效果,是一种具有良好材料特性的组织工程材料。[ Objective] To manufacture collagen typeI/VEGF (vascular endothelial growth factor, VEGF) slow-release timber biomaterial, investigate the slow-release effectiveness of this material, and find a new composition material for the tissueengineering of bone-tendon tissue. [ Method] This material was surveyed in respect with mechanic, aperture, uhraviolet absorp-ion maximum light spectrum, porosity, water content, fourier infrared spectrum,and tissue compatibility with VEGF, and utilized the biological method load the VEGF to collagen type Ⅰ, and the slow-release ability combined material was prepared. [ Resuit]The collagen type I sponge had the ability to encapulate the VEGF. And VEGF was slowly eleased from collagen type I sponge. This material had good biocompatibility with VEGF. [ Conclusion ] The collagen typeⅠ has desirable aperture and advisabie biomechanics. This material will be a good material for the bone-tendon tissue engineering.
