摘  要:目的观察IL-6基因启动子区-174G/C及-572C/G多态性在中国汉族人群中的分布频率与T2DM患者大血管病变(DVC)的遗传易感性的关系。方法应用聚合酶链反应-限制性片段长度多态性分析(PCR-RFLP)方法检测480例患者(DM组120例,DVC组160例,对照组200例)-174位点、-572位点多态性。结果 IL-6基因启动子区-174G/C多态性中,携带C等位基因的个体患DVC风险为G等位基因的1.8271倍(95%CI:1.1785~2.8327,P<0.01)。-572C/G多态性在DM+DVC组与对照组比较,差异无统计学意义(P>0.05)。结论 IL-6-174G/C基因多态性与DVC的发病具有相关性。C等位基因可能是DVC的遗传易感基因。Objective To study association between the polymorphisms in the promoter of interleukin-6 (IL-6) gene -174G/C and -572C/G macrovascular disease (MA) in type 2 diabetes mellitus (T2DM) in Chinese Han population. Methods Using the PCR-restriction fragment length polymorphism method, two IL-6 promoter polymorphism genes (-174G/C, -572C/G) were studied in 480 subjects including 120 diabetes without macrovascular disease, 160 diabetes with macrovascular disease and 200 controls. Results For -174G/C polymorphism in the promoter of IL-6 gene, the incidence of macrovascular disease in type 2 diabetes in the individuals with C allele was significantly increased in comparison with the carriers of G allele(OR= 1. 8271,95% CI: 1. 1785-2. 8327, P^0. 01). There was no statistical difference between DM+ DVC and control groups for -572C/G polymorphism. Conclusion These results suggest that the -174G/C polymorphism in the promoter of IL-6 gene is associated with macrovascular disease in type 2 diabetes. The C allele in -174G/C of IL-6 gene may contribute to the predisposition gene locus for macrovascular disease in type 2 diabetes.
