摘 要:采用SDS-PAGE法和近红外反射光谱法检测254份品种的HMW-GS和其中188份的沉淀值、蛋白质和湿面筋含量,研究甘肃春小麦育成品种和地方品种HMW-GS变异及品质效应。结果表明,共检测到21种亚基变异,46种亚基组成形式,育成品种亚基变异和组成形式不及地方品种丰富。Glu-1不同位点上,育成品种1、7+8和2+12分别为各自位点优势亚基(69.%、81.8%和86.0%),地方品种null、7+8和2+12为各自位点优势亚基(77.4%、88.7%和76.7%);育成品种1、14+15、5+10对蛋白质含量、沉淀值和湿面筋含量的效应值最高,地方品种2*、7+8和5+10对蛋白质含量的效应值最高,1、7+8和5+10亚基沉淀值和湿面筋含量的效应值最高。就优质亚基出现的频率而言,育成良品种Glu-A1位点品质改良效果较好。Allelic variations of HMW-GS in 254 Gansu spring wheat landraces were studied by SDS-PAGE,and the quality including sedimentation,protein and wet gluten contents of 188 landraces were analyzed by near-infrared reflectance spectroscopy(NIRS) in this experiment.The results showed that 21 subunit types and 46 subunit compositions were detected in the Glu-1 loci,which implied that the landraces had much more diversities than that of the improved varieties.The subunits composition of 1(69.%),7+8(81.8%),2+12(86.0%) was the major type in the improved varieties,as well the subunits composition of null(77.4%),7+8(88.7%),2+12(76.7%) was the dominate combination in the landraces.The effect of 1,14+15,5+10 composition on quality of protein content,sedimentation and wet gluten content was the highest in the improved varieties,but the subunit combination 2*,7+8,5+10 on quality of protein content was the highest in the landraces,and 1,7+8,5+10 on quality of sedimentation and wet gluten content was the highest.The frequency of fine subunit at Glu-A1 locus was higher than that at Glu-B1 and Glu-C1 loci in the improved varieties.
