摘 要:目的分析广西近几年来分离的流行性乙型脑炎病毒(乙脑)与减毒活疫苗株(SA14-14-2株)之间在E基因区段核苷酸及氨基酸差异。方法广西6株乙脑病毒株E基因序列由本实验室测定获得,减毒活疫苗株和131V株序列从GenBank中下载获得,应用Clustal X(1.8)、DNASTAR等生物软件进行分析。结果广西分离株与SA14-14-2株之间的核苷酸同源性在87.4%~88.1%之间,氨基酸同源性在96.8%~97.2%之间。SA14-14-2株与广西分离株之间共存在19个位点的差异,其中E107、E129、E138、E176、E177、E222、E244、E264、E279、E315、E327、E366、E439、E447处所有广西分离株与SA14-14-2株存在共同差异。结论广西分离的乙脑病毒与减毒活疫苗株在E基因的部分氨基酸位点存在差异,但均不处在影响病毒生物学活性的关键位点。使用减毒活疫苗株SA14-14-2株能保护新分离株引起的感染。Objective To analyze the difference of nucleotide and deduced amino acid sequences of E gene between the Japanese encephalitis(JE) virus strains isolated in Guangxi and the attenuated vaccine strain SA14-14-2.Methods E gene sequences of six JE virus strains isolated in Guangxi were sequenced in our laboratory,and the sequences of the attenuated vaccine strain SA14-14-2 and 131V were obtained from GenBank.The bioinformatics softwares such as Clustal X(1.8) and DNASTAR were used to analyze the nucleotide and deduced amino acid sequences.Results The results showed that the nucleotide sequence and deduced amino acid sequence homologies between Guangxi isolated strains and the attenuated vaccine strain SA14-142 were up to 87.4%,and the 96.8%,respectively.Compared with the attenuated vaccine strain SA14-14-2,there were nineteen amino acid mutations in E gene of all Guangxi isolated strains.There were fourteen common mutations at E107,E129,E138,E176,E177,E222,E244,E264,E279,E315,E327,E366,E439,and E447 sites between all isolated strains and SA14-14-2.Conclusion There are some differences between SA14-14-2 and JE viruses isolated from mosquitoes and human in Guangxi.But these mutations are not in the important sites affecting the biological charactetistic of the viruses.Using attenuated vaccine can protect human from infection by these JE viruses.
