摘  要:目的探讨滋阴清热的玉女煎治疗自发性2型糖尿病GK大鼠胰岛自噬基因Beclin表达的变化。方法 GK大鼠分为模型对照组和观察组,同品系的Wistar大鼠作正常对照组;观察组用玉女煎灌胃,另2个对照组用生理盐水灌胃,每天1次,连续4周。取胰腺,用免疫组化方法检测胰岛自噬基因Beclin的表达;电子显微镜观察胰岛β细胞自噬的形态学变化。结果玉女煎组胰岛自噬基因Beclin的表达和GK对照组相比显著降低,而和Wistar大鼠正常对照组之间没有显著差异;电镜下可见对照组GK大鼠胰岛β细胞内有较多的自噬体。结论自噬参与了2型糖尿病的病理生理过程,玉女煎可降低胰岛β细胞自噬基因Beclin的表达。Objective To explore the changes of islet autophagy gene beclin expression of spontaneous diabetic GK rat treated by Yunvjian which has the effect of nourishing yin and eliminating heat.Methods The GK rats were divided into model control group and observation group,and Wistar rats of same strain were chosed as normal control group.The observation group received Yunvjian and the two control groups received normal saline by gastric perfusion for 4 weeks.Then pancreas tissue sample was collected,and the expression of islet autophagy gene beclin was detected by immunohistochemical method and the morphological changes of islet β cells were observed by using electron microscope.Results The expression of islet autophagy gene beclin was decreased significantly in Yunvjian group compared with model control group,but there was no significant difference between Yunvjian group and normal control group.Many autophagic bodies were found in islet β cells of GK rats in model control group by electron microscope.Conclusion Autophagy may participate in the pathophysiological process of type 2 diabetes mellitus,and Yunvjian could reduce the expression of islet autophagy gene beclin.
