摘 要:为研究大鼠促卵泡素(FSH)分泌的受体后信号转导机制,将GTH细胞用PMA或H7处理后,用GnRH脉冲刺激,再用实时荧光定量PCR方法测定细胞FSH-βmRNA表达的Ct值,并与空白对照组比较。结果表明,PMA能显著降低GTH细胞FSH-βmRNA表达的Ct值,H7则曾能显著升高,说明GTH细胞PKC活性显著影响FSH-βmRNA的表达,FSH-βmRNA的表达随着PKC活性的升高而显著升高,随着PKC活性的降低而显著降低。因此,PKC-Ca2+是GnRH脉冲刺激所引起的FSH-βmRNA表达的受体后的信号转导途径。This article is to analyze the post-receptor signal transduction mechanism of the secretion of follicle stimulating hormone(FSH) in rats.The gonadotropic hormone(GTH) cells was stimulated by gonadotrophin hormone-releasing hormone(GnRH) after the activity of protein kinase C(PKC) in them was treated by phorbol 12-myristate 13-acetate(PMA) or 1(5-isoquinoline sulfonyl)-2-methyl piperazine dihydroch loride(H7).Ct of FSH-β mRNA was detected by real time quantitative PCR(RTqPCR),which was compared with the control group.The result showed that PMA can significantly reduce the Ct of FSH-β mRNA in GTH cells,while H7can significantly improveit.The activity of PKC in GTH cells influences the expression of FSH-β mRNA significantly,the expression of FSH-β mRNA rises with the rise of PMA and falls with the falling of H7.The conclusion is drawn that PKC-Ca ^2+ is the post-receptor signal transduction pathway that the cells stimulated by GnRH impulse express FSH-β mRNA.
