摘  要:目的 研究胰岛素样生长因子I受体 (IGF IR)基因表达对脐血T淋巴细胞功能的影响及其临床意义。方法 利用脂质体将IGF IR反义寡核苷酸 (ASON)转入脐血淋巴细胞 ,通过同位素掺入法了解淋巴细胞增殖功能 ,采用逆转录聚合酶链反应技术检测IGF IR、IL 2、IL 4和干扰素 (IFN)r的mRNA表达水平。结果 通过IGF IRASON可抑制脐血淋巴细胞IGF IRmRNA的表达约 5 0 % ,同时淋巴细胞的增殖功能较单纯PHA刺激也下降 5 0 %左右 ;细胞因子IL 2、IL 4、IFNr的mRNA水平也有不同程度的降低 ,其中IL 2和IFNr的mRNA水平经IGF IRASON作用后较单纯PHA刺激相比降低幅度约为 30 %~ 5 0 % ,IL 4mRNA降低幅度约为 2 0 %~ 30 %。结论 IGF IR参与脐血淋巴细胞的增殖和细胞因子的产生 ,提示在维持细胞免疫功能和调节婴儿时期TH1 TH2应答相对平衡的过程中IGF IR可能具有重要意义。Objective To study the role of insulin like growth factor type I receptors (IGF IR) in the modulation of cord blood T lymphocyte functions and its clinical significance. Methods The antisense oligonucleotide was used to inhibit the expression of IGF IR mRNA of cord blood lymphocyte. By using the reverse transcriptase polymerase chain reaction and 3H thymidine uptake technique, the mRNA expressions of IGF IR, IL 2, IL 4, IFN γ as well as the proliferation of lymphocyte were detected. Results A 50% reduction in IGF IR mRNA following exposure to antisense oligonucleotide was observed. The proliferation of T cell to mitogens was significantly reduced about 50%. The mRNA levels of IL 2, IL 4 and IFN γ were also markedly decreased after using IGF IR antisense oligon ucleotide. Compared with the expore to PHA, the mRNA levels of IL 2 and IFN γ decreased by 30%~50%, and IFN γ decreased 20%~30%. Conclusion IGF IR was involved in the process of cord blood lymphocyte proliferation and the production of cytokines. It might play an important role in maintaining cell immune functions and T H1/T H2 relative balance in infants.
