摘  要:【目的】对Ogura胞质雄性不育系及恢复系进行遗传差异分析,为生产上利用Ogura胞质不育系统选育强优势甘蓝型油菜杂交组合提供参考。【方法】利用分布于连锁群上的76对SSR标记分析79份甘蓝型油菜Ogura细胞质雄性不育系及恢复系材料间的差异并进行聚类分析。【结果】在76对SSR标记引物(每条连锁群4对SSR标记)中可筛选出38对标记;在79份材料中,38对SSR标记共扩增到123条清晰条带,扩增片段大小为100~400bp;获得89个多态性位点,材料间遗传相似系数为0.52~0.99。聚类分析结果表明,在相似系数0.68处,可将79份材料分为A、B、C、D、E5个类群,92.4%的材料可归入A、B两类,其中72.1%的恢复系材料归入A类,72.8%的不育系材料归入B类。A类和B类又可在相似系数0.74处分别分为A1、A2、A3、A4、A5、A6、A7及B1、B2、B3、B411个亚群。【结论】SSR标记可用于甘蓝型油菜Ogura细胞质雄性不育系及恢复系的分类鉴定。[Objective]The present study was conducted to analyze genetic diversity of Ogura male sterile line and restorer line to provide reference for breeding strong heterosis hybrids of Brassica napus using Ogura cytoplasm sterility.[Method]Seventy six pairs of SSR primers distributed on linkage map were used for variance analysis and cluster analysis among 79 Ogura male sterile line and restorer line of Brassica napus.[Result]Among 76 pairs of SSR primers,38 were selected to amplify 123 clear bands ranging from 100-400 bp in 79 accessions,and 89 polymorphic loci were obtained.Genetic similarity ranged between 0.52-0.99.Seventy-nine B.napus lines were clustered into five groups of A,B,C,D and E at coefficient of 0.68.About 92.4% of total lines were classified into groups A and B,and 72.1% of the male sterile lines were classified into group A,72.8% of the restorer lines were classified into group B.Groups A and B could also be divided into A1,A2,A3,A4,A5,A6,A7 and B1,B2,B3,B4 sub-groups at coefficient of 0.74,respectively.[Conclusion]SSR marker can be used for identifying classification Ogura male sterile line and restorer line.
