东方巴贝斯虫分泌性抗原的免疫保护作用研究

摘　　要：体外培养获得的东方巴贝斯虫 (BabesiaorientalisLiuZhonglingetal.1997)分泌性抗原经离心浓缩等处理后用于免疫预防作用的研究。 10头从东方巴贝斯虫病非疫区购回的 1岁左右的健康水牛犊分为 3组 ,免疫组 4头牛 ,用分泌性抗原和弗氏完全佐剂混合后间隔二周免疫二次 ,佐剂对照组 2头牛 ,用弗氏完全佐剂间隔二周注射二次。空白对照组 4头牛不作处理。二免后一个月取液氮保存的东方巴贝斯虫含虫血进行攻击 ,结果发现 :免疫组牛获得了良好的免疫保护 ,除出现红细胞数及血红蛋白含量的下降外 ,无其它明显可见的病理反应 ,佐剂对照组二头牛均出现了虫血症并死亡一头。空白对照组 4头牛全部出现了明显的虫血症 ,且均以死亡告终。试验过程中对Ab和T淋巴细胞的检查表明该免疫原诱导产生了体液和细胞免疫反应。本试验的结果说明了东方巴贝斯虫体外培养分泌性抗原具有良好的免疫保护作用。WT5BZ] Culture derived Babesia orientalis exoantigens were used to vaccinate buffalo calves.10 one year old health buffalo calves purchased from babesiosis nonepidemic areas were divided into three groups.Vaccinated group(4 buffalo calves)received two subcutaneous immunizations with a two week interval of exoantigens plus Freunds adjuvant,adjuvant control group(2 buffalo calves)received two injections of adjuvant alone.Control group(4 buffalo calves)didnt receive any injection.One month after the second immunization,all animals were challenged subcutaneously with 2×10 8 virulent Babesia orientalis organisms.The results showed that exoantigens of Babesia orientalis could induce a high degree of protection against virulent Babesia orientalis challenge exposure.In vaccinated group,all animals only developed a slight decrease in Hb and RBC.In adjuvant group,one animal died,the other showed parasitaemia.All animals of the control group showed typical clinical symptoms and died between 11 and 16 days after challenge.Vaccination elicited both humoral and cell mediated immune responses.
