摘  要:目的了解运动神经元和神经干细胞诱导分化所得胆碱能神经元间miR-126和miR-31的差异表达情况,并以此来探讨两种细胞之间的差异。方法应用ABI公司的TaqMan MicroRNA Assays real-time PCR技术,观察miR-126和miR-31在运动神经元与神经干细胞分化所得胆碱能神经元中的表达情况。结果 miR-126在神经干细胞分化所得胆碱能神经元中的表达是在运动神经元中的0.002倍(P<0.05)。miR-31在神经干细胞分化所得胆碱能神经元中的表达是在运动神经元中的56.444倍(P<0.05)。结论 miR-126和miR-31在运动神经元与神经干细胞分化所得胆碱能神经元中的表达存在差异,对二者预测靶基因参与的生物学过程分析,暗示两种细胞可能在信号传导和发育上存在有差别。Objective To study the differential expressions of miR-126 and miR-31 gene between motor neurons and cholinergic neurons derived from neural stem cells,and learn the different between motor neurons and cholinergic neurons. Methods ABI TaqMan MicroRNA Assays real-time PCR was used to measure the expressions of miR-126 and miR-31 in motor neurons and cholinergic neurons derived from neural stem cells. Results The expression of miR-126 in cholinergic neurons derived from neural stem cells was 0.002-fold of that in motor neurons(P0.05),While the expression of miR-31 in cholinergic neurons derived from neural stem cells was 56.444-fold of that in motor neurons(P0.05). Conclusion The expressions of miR-126 and miR-31 are different between motor neurons and cholinergic neurons derived from neural stem cell.The biological process,in which miR-126 and miR-31 are involved about forecasting turget genes,suggests that the two kinds of cell may be different in signal conduction and development.
