摘 要:目的探讨布地奈德气雾剂对哮喘气道重塑大鼠气道平滑肌细胞(airway smooth muscle cell,ASMC)中MMP-9(金属基质蛋白-9)及TIMP-1mRNA(组织抑制因子-1)表达的影响。方法将60只wistar大鼠按照随机分组原则分为正常组、模型组及治疗组。模型组采用Wistar大鼠哮喘模型制作方法制作哮喘气道重塑大鼠模型;HE染色图像分析测量各组大鼠肺组织中气道壁面积(Wat)及平滑肌层面积(Was),并用气道基膜周长(Pbm)进行标准化;原代培养各组大鼠ASMC;实时定量PCR检测比较各组大鼠ASMC中MMP-9及TIMP-1mRNA表达含量,用SPSS 17.0统计软件进行统计学分析,组间比较采用单因素方差分析,P<0.05为差异有统计学意义。结果模型组大鼠肺组织中Wat/Pbm及Was/Pbm明显较正常对照组增加,治疗组大鼠肺组织中Wat/Pbm及Was/Pbm较正常对照组增加,但较模型组明显降低,差异均有统计学意义(P<0.05)。模型组大鼠ASMC中MMP-9mRNA表达较正常对照组增加,差异有统计学意义(P<0.05);但治疗组大鼠ASMC中MMP-9mRNA表达较模型组减少,差异有统计学意义(P<0.05);但较正常对照组大鼠ASMC中含量增加(P<0.05)。模型组大鼠ASMC中TIMP-1mRNA表达较正常对照组降低;但治疗组大鼠ASMC中TIMP-1mRNA表达较哮喘组增加,但较正常对照组降低(P<0.05)。结论布地奈德通过降低哮喘大鼠ASMC中MMP-9mRNA,增加TIMP-1mRNA表达,从而减轻哮喘大鼠ASMC细胞间质增厚。Objective To investigate the effect of budesonide on the expression of MMP-9 and TIMP-1 mRNA in asthmatic rats ASMC. Methods Sixty wistar rats were randomly divided into 3 groups: control group,model group and treatment group.In the model group,airway remodeling was made with the conventional method.HE staining was used to measure the wat and was in different groups;primary culture ASMC of different groups;real-time PCR analyzed the expression of MMP-9 and TIMP-1 mRNA in ASMCs of different groups.SPSS 17.0 and One-way ANOVA were used to analyze the difference among groups.P0.05 was considered significant. Results Wat/Pbm and Was/Pbm in model group increased when compared with those in normal group;Wat/Pbm and Was/Pbm in the treatment group was higher increased than in the control group but lower than in the model group.The expression of MMP-9 mRNA in the treatment group was lower than in the model group but higher than in the control group,with significant differences.The expression of TIMP-1 mRNA in the treatment group was lower than in the control group,but higher than in the model group,with significant differences. Conclusion Budesonide alleviates airway remodeling in asthma by decreasing the expression of MMP-9 mRNA and increasing the expression of TIMP-1 mRNA in ASMC of asthmatic rats.
