摘  要:目的探讨不同时辰电针对氯胺酮滥用成瘾戒毒治疗的形态学基础。方法将56只SD大鼠随机分为正常对照组、生理盐水(10ml/kg)组、氯胺酮(100mg/kg)组、氯胺酮加电针组(分23:00子时、05:00卯时、11:00午时、17:00酉时电针组)。按上述设定剂量,经腹腔注射给药,每天一次,连续7天,氯胺酮加电针组于给药一周后分别在四个时辰给予低频(2Hz)电针交替刺激"足三里"与"三阴交"穴,持续30min/次/日,连续一周,采用免疫组织化学染色方法显示腹侧苍白球酪氨酸羟化酶、c-fos的表达。结果与正常组和生理盐水组相比较,氯胺酮组腹侧苍白球酪氨酸羟化酶、c-fos表达明显增强(P<0.01)。与氯胺酮组相比较,午时、酉时电针组可使酪氨酸羟化酶、c-fos表达明显减弱(P<0.01),而子时、卯时电针组无明显变化(P>0.05)。结论氯胺酮滥用可以增强酪氨酸羟化酶、c-fos在腹侧苍白球的表达,午时、酉时电针具有逆转作用。Objective To explore the morphological basis of electro-acupuncture at different time on the treatment of ketamine abuse. Methods 56 Sprague-Dawley rats were randomized into normal control group,saline group(10ml/kg),ketamine group(100mg/kg) and ketamine﹢electroacupuncture group(divided into 05:00,11:00,17:00 and 23:00 subgroups).Each group was intraperitoneally given the above dosage once a day for 7 days.Electroacupuncture of 2HZ was applied to Zusanli and Sanyinjiao acupoints,30 min a day for 7 days.Immunohistochemistry was used to study the expression of Tyrosine hydroxylase and c-fos. Results Compared with those of normal control and NS groups,Tyrosine hydroxylase and c-fos expression was enhanced in Ventral Pallidum of the ketamine group(P0.01).Compared with the ketamine group,the 11:00 and 17:00 electroacupuncture groups weak ened Tyrosine hydroxylase and c-fos expression(P0.01),but the 23:00 and 05:00 electroacupuncture groups showed no obvious difference(P0.05). Conclusion Ketamine abuse can promote Tyrosine hydroxylase and c-fos expression in Ventral Pallidum,and 11:00 and 17:00 electroacupuncture can reverse this effect.
