摘 要:为了能在分子生物学基础上进行资源保护和提高综合开发利用价值,对黄山市几种资源植物进行了初步的分子生物学研究。通过3种不同方法(普通CTAB法、改良CTAB法和SDS法)对黄山贡菊、艾草、玉叶金花、柳叶蜡梅和秤锤树这5种黄山市资源植物,进行叶片基因组总DNA提取方法的研究,并用紫外分光光度法和1%琼脂糖凝胶电泳法分别检测所得产物DNA的纯度和完整性。结果表明:对于黄山贡菊、艾草和玉叶金花基因组总DNA的提取采用改良CTAB法所得提取效果最佳;对于柳叶蜡梅以普通CTAB法提取效果最佳;对于秤锤树以SDS法提取效果最佳。琼脂糖凝胶电泳结果显示完整度较好,能够用于进一步的分子生物学研究。Abstract: In order to protect the natural resources based on molecular biology and appreciate the comprehensive development and utilization value, we carried out a pilot study on molecular biology of several resource plants in Huangshan. The abstraction methods of genomic DNA from the leaves of several resource plants in Huangshan, such Mussaenda pubescens Ait. f., as Dendranthema morifolium (Ramat) Tzvel. cv. Gongju, Artemisia argyi, Chimonanthus salicifolius S.Y.H and Sinojackia xylocarpa Hu had been studied. The three methods, usual CTAB, modified CTAB and SDS were compared. Then using ultraviolet specrophotometer and 1% agarose gel electrophoresis tested the purity and integrality of the genomic DNA extraction. The experiment results indicated that, the modified CTAB method obtained better effect to extract the genomic DNA from Dendranthema morifolium (Rama( pubeseens Ait. f. The usual CTAB method obtained bettert) Tzvel. cv. Gongju, Artemisia argyi and Mussaenda effect extract the genomic DNA from Chimomanthus salicifolius S.Y.H. For Sinojackia xylocarpa Hu, SDS method obtained better effect. The DNA distracted by the three methods showed better integrity by 1% agarose gel electrophoresis. So the DNA extraction could be used for molecular biology analysis.
