摘  要:目的应用肠外翻模型研究葛根芩连汤(葛根、黄芩、黄连和炙甘草)及其不同配伍组中4个生物碱类成分在大鼠不同肠段的吸收特征。方法采用肠外翻模型,考察葛根芩连汤中黄连碱、药根碱、小檗碱、巴马汀4个生物碱类成分的肠吸收机制;同时考察不同肠段、药物浓度等因素对它们吸收的影响并比较各组分在全方和不同配伍组给药时的吸收变化情况。结果各组分在不同肠段的吸收均为线性吸收,符合零级吸收速率。药根碱在各肠段的吸收方式除被动扩散外还存在主动转运。小檗碱、巴马汀的吸收速率常数(Ka)均随剂量的增加而增加,显示它们为被动扩散。黄连碱在空肠中的吸收表现为主动转运;而在十二指肠、回肠、结肠中表现为被动扩散。肠道不同部位的吸收实验表明,全方中小檗碱、巴马汀的最佳吸收部位为空肠;黄连碱、药根碱的最佳吸收部位为结肠。结论比较不同配伍组的Ka,4个生物碱成分在全方中吸收最好,其他三个配伍组的吸收没有显著性差异。AIM To use rat everted gut sacs to investigate the intestinal absorption characteristics of alkaloids in Gegen Qinlian Decoction (Puerariae lobatae Radix, Scutellariae Radix, Coptidis Rhizoma, Glycyrrhizae Radix et Rhizoma praeparata cure melle) and different compatibilities. METHODS In the research, the intestinal absorption mechanism of the active constituents including coptisine, jateorhizine, berberine and palmatine in Gegen Qin- lian Decoction was studied. And then the influences of different intestinal segments, different drug concentrations on the absorption of constituents were discussed. Finally, the absorption characteristics of the active compounds in different compatibility were compared. RESULTS The absorption mode of the four compounds corresponded to the zero order rate process. The mechanism for absorption of jateorhizine across intestinal sacs was passive diffu- sion, and active transportation also existed. The Ka of berberine and palmatine increased along with the raised dos- age, and the two compounds displayed the passive absorption. The mechanism for absorption of coptisine was the active transport in the jejunum and belonged to the passive absorption in the duodenum, colon and ileum. The results of experiment of intestinal absorption in different intestinal segments showed that it was from Gegen Qinlian Decoction that jejunum was the best absorption site of berberine and palmatine and that colon was the best one of coptisine and jateorhizine. CONCLUSION In the comparison with Ka of different compatibilities, the absorption of the four alkaloids from Gegen Qinlian Decoction is ideal.
