摘 要:目的比较并评价四逆汤的传统制备法与《中国药典》2010年版一部所载制备方法。方法采用RP-HPLC测定并比较两种方法制备的四逆汤中甘草酸、新乌头碱、乌头碱、次乌头碱、6-姜辣素的含有量及干膏量。甘草酸色谱柱为Krosmasil C18柱(150 mm×4.6 mm,5μm),以甲醇-0.2 mol/L醋酸铵溶液(65∶35,含0.5%冰醋酸)为流动相,检测波长为250 nm;双酯型二萜类生物碱(DDA)色谱柱为Symmetry Shield RP18柱(250 mm×4.6 mm,5μm),以甲醇(A)-0.05%三乙胺水溶液(B)梯度洗脱,检测波长为237 nm;6-姜辣素色谱柱为Symmetry Shield RP18柱(250 mm×4.6 mm,5μm),以乙腈-甲醇-水(40∶5∶55)为流动相,检测波长为280 nm。结果按传统方法制备四逆汤含甘草酸、6-姜辣素的量和干膏量分别是按中国药典方法制备四逆汤的3.6倍、1.7倍和5倍,新乌头碱、乌头碱和次乌头碱的含有量总和相近。结论从化学成分的角度,初步揭示四逆汤的传统制备方法与中国药典方法相比存在一定差别。AIM To compare and evaluate the different preparation methods between the traditional method and Chinese Pharmacopoeia (2010, I) method on Sini Decoction. METHODS The contents of glycyrrhizic acid, mesaconitine, aconitine, hypaeonifine and 6-gingerol were determined by RP-HPLC in Sini Decoction prepared with different processes, and the yield of dry extract was estimated. The chromatographic conditions were as fol- lows, for glycyrrhizic acid: Kromasil C18 column ( 150 mm×4.6 mm, 5μm) with mobile phase of methanol -0.2 mol/L ammonium acetate (65 : 35, with 0. 5 % glacial acetic acid) and the detection wavelength at 250 nm, for diester diterpenoid alkaloids : Symmetry Shield RP,8 (250 mm ×4.6 mm, 5 μm) with mobile phase of methanol (A) - 0. 05 % triethylamine ( B), gradient elution and the detection wavelength at 237 nm, for 6-gingerol : Symmetry Shield RPls(250 mm x4.6 ram, 5 txm) with mobile phase of acetonitrile-methanol-warter (40 : 5 : 55) and the detection wavelength at 280 nm. RESULTS The contents of glycyrrhizic acid, 6-gingerol and the yield of dry extract in Sini Decoction prepared with traditional method were 3.6, 1.7 and 5 times the contents in the decoction prepared with the method in Chinese Pharmacopoeia. The total contents of three kinds of diester diterpenoid alkaloids were the same in Sini Decoction prepared with different methods. CONCLUSION From the perspective of chemical composition, it is initially revealed that there are differences between the traditional process on Sini Decoction and the method reported in Chinese Pharmacopoeia.
