摘 要:目的探讨格列美脲在正常大鼠和糖尿病大鼠体内的药动学差异以及黄芪注射液对其体内药动学的影响。方法正常Wistar大鼠10只,分为A组和B组;经链脲佐菌素造模的糖尿病大鼠10只,分为C组和D组,每组5只。A组和C组灌胃给予格列美脲混悬水溶液100 mg·kg^(-1),B组和D组静脉注射黄芪注射液8 mg·kg^(-1),5 min后灌胃给予格列美脲混悬水溶液100 mg·kg^(-1)。于不同时间点股动脉采集血样,处理后经HPLC法测定格列美脲的血药浓度,绘制血药浓度-时间曲线,采用DAS 2.0软件处理数据。结果与A组相比,C组格列美脲的AUC增高,CL降低(P<0.01)。与B组比较,D组格列美脲的AUC增高,CL降低(P<0.01)。B组格列美脲的AUC高于A组,CL低于A组(P<0.05)。D组格列美脲AUC高于C组,CL低于C组(P<0.01)。结论糖尿病大鼠格列美脲CL降低,黄芪注射液可延缓格列美脲在正常大鼠和糖尿病大鼠体内的消除,2药合用时应适度降低格列美脲的用量。AIM To study the pharmacokinetics of glimepiride and effects of co-administration Huangqi injection on the pharmacokinetics of glimepiride in normal and diabetic rats. METHODS Ten normal rats were randomly divided into group A and group B (5 in each), and ten diabetic rats induced by streptozotocin were randomly divided into group C and group D (5 in each). The rats in the group A and group C were administered orally of glimepiride aqueous solution at a single dose. The rats in the group B and group D were administered orally of glimepiride aqueous solution (100 mg .kg-1) after rats were intravenously administered with Huangqi injection (8 mg'kg-~) for 5 min. The blood samples were collected at different time, and the concentrations of glimepiride in plasma were determined by HPLC. According to the curve of concentration-time, the software of DAS 2.0 was used to process the pharmacokinetic parameters. RESULTS Compared with that in the group A, the AUC of glimepiride in the group C has increased, and the CL has decreased (P 〈 0.01). Compared with that in the group B, the AUC of glimepiride in the group D has increased, and the CL has decreased (P 〈 0.01). The AUC of glimepiride in the group B has significantly increased and the CL has significantly decreased when compared with that in the group A (P 〈 0.05). The AUC of glimepiride in the group D has significantly increased and the CL has significantly decreased when compared with that in the group C (P 〈 0.01). CONCLUSION Huangqi injection could decrease the CL of glimepiride in the normal and diabetic rats, which suggested the dose of glimepiride should be reduced when co-administering with Huangqi injection.
