苦参碱与其联合顺铂对U14荷瘤小鼠抑制作用的研究

摘　　要：目的探讨苦参碱及苦参碱联合顺铂对小鼠移植性宫颈癌的抑制作用,分析苦参碱是否能减少顺铂的副作用。方法建立U14荷瘤小鼠模型,随机分成8组(每组10只):阴性对照组,顺铂组,苦参碱高、中、低剂量组,顺铂联合苦参碱高、中、低剂量组。以抑瘤率为指标,观察各组药物对荷瘤小鼠肿瘤生长的抑制作用;以小鼠的一般状况及血清肌酐、尿素氮为指标,观察联合用药是否能减少顺铂治疗的副作用。结果苦参碱高、中剂量组和联合用药各组对U14实体瘤均有不同程度的抑制作用,抑瘤率均>30%且苦参碱高剂量组和联合用药各组瘤重较阴性对照组显著减少(P<0.01),苦参碱中剂量组瘤重较对照减少,差异有统计学意义(P<0.05)。联合用药各组荷瘤小鼠一般情况较顺铂组好,联合高、中剂量组肌酐、尿素氮水平较顺铂组低(P<0.05)。结论苦参碱具有抗肿瘤作用,与顺铂联合使用,在增加抑瘤作用的同时,可适度减少顺铂的副作用。Objective To study the inhibitory effects of Matrine and Matrine plus cisplatin on transplanted cervical cancer of mice,and analyse if Matrine can reduce the side-effect of the treatment with cisplatin on cervical cancer.Methods To create U14 tumor model and randomly divided into 8 groups（10 mice each）which included negativecontrol group,cisplatin group,high dose group of Matrine,medium dose group of Matrine,low dose group of Matrine and the aforementioned three dose groups of Matrine plus cisplatin.The inhibitory effects of these groups of drugs on tumor growth were evaluated by calculating the reduced rate of tumor growth.By using Cr,BUN and the general condition of mice as index which was in order to observe if drug combination can reduce the side-effect of cisplatin.Results The groups of high,medium dose Matrine and three dose Matrine plus cisplatin had inhibitory effects on tumor growth of U14 tumor-bearing mice,the inhibition rate of all these groups were greater than 30% and the tumor weight was lower in medium dose group of Matrine（P 0.05）,and statistically significant lower in cisplatin group,high dose group of Matrine and three combined treatment groups（P 0.01） that compared with the tumor weight of negative control group.The value of Cr and BUN in two groups which were high,medium dose Matrine plus cisplatin was lower than that was in cisplatin group（P 0.05）,the general condition of these groups above were better than what was in cisplatin group.It means Matrine combined treatment could appropriately reduce the toxicity of cisplatin.Conclusion Matrine has anti-tumor effect.At the same time,if combined with cisplatin,Matrine will have not only the antitumous effect but also the capability of reducing the toxicity of ciaplatin treatment.
