摘 要:本研究旨在通过对内蒙古黑猪成年母猪后肠木聚糖降解菌的分离、筛选及鉴定,并对比研究筛选菌和瘤胃液对饲草纤维降解的作用,获得可高效降解木聚糖的菌株。以木聚糖为唯一碳源,根据筛选平板上菌株透明圈大小和发酵液中木聚糖酶活性分离得到了2株活性较高的木聚糖降解菌(分别命名为ZF和MZ)。经形态学观察和16S rDNA核苷酸序列分析表明,菌株ZF和MZ分别为兼性厌氧产黄纤维单胞菌(Cellulomonas flavigena)和类芽孢杆菌(Paenibacillus sp.)。饲草纤维降解结果表明:瘤胃液对苜蓿和青贮玉米的中性洗涤纤维(NDF)和酸性洗涤纤维(ADF)降解率均高于筛选菌;菌株ZF和MZ对3种饲草NDF和ADF降解率均有随发酵时间延长而增加的趋势,其中菌株ZF降解羊草ADF和NDF的效果等同甚至优于瘤胃液。结果提示,从成年母猪后肠中筛选的木聚糖降解菌株ZF和MZ可有效降解饲草中的纤维类物质,可作为生产饲用木聚糖酶的候选菌株。This experiment was conducted to study the isolation,screening and identification of efficient xylan-degrading bacteria from the hindgut of high roughage-resistance Inner Mongolia black adult sows,and to compare the effects of screening bacteria and rumen liquid on neutral detergent fiber(NDF) and acid detergent fiber(ADF) degradation of forages(alfalfa,Chinese wildrye and silage corn).Using xylan as the sole carbon source,two xylan-degrading bacteria named ZF and MZ were isolated by screening with the diameter of clearing zone and xylanase activity.Strains of ZF and MZ were identified as Cellulomonas flavigena and Paenibacillus sp.,respectively,which were facultative anaerobe strains,by morphology observation and 16S rDNA sequence analysis.The degradation results showed that the degradabilities of NDF and ADF in alfalfa and silage corn by rumen fluid were higher than those by strains of ZF and MZ.With fermentation time prolonging,the degradabilities of NDF and ADF in Chinese wildrye by strain ZF and MZ had a tendency to increase,and the degradabilities of NDF and ADF in Chinese wildrye by strain ZF were equal or higher than those by rumen fluid.In conclusion,xylana-degrading bacteria ZF and MZ isolated from the hindgut of adult sows have efficient ability to degrade fiber;therefore,they can be used as candidate strains for xylanase production.
