摘 要:目的鉴定高性能战斗机飞行员的PERIOD3(PER3)基因型频率,并分析其与睡眠分型的关系。方法获取104名高性能战斗机飞行员的外周静脉血样本各2m1,提取基因组DNA样本。应用PCR扩增基因片段,鉴定个体的PER3基因型。根据Horne-Ostberg生理时钟问卷调查,进行睡眠类型分类。结果104名高性能战斗机飞行员的PER3基因型频率为:PER3^4/4基因型频率占70.2%(73/104);PER3^4/5基因型频率占26.9%(28/104);PER3^5/5频率占2.9%(3/104)。高性能战斗机飞行员与普通汉族人PER3基因型频率比较,差异无统计学意义(X^2=2.737,P〉0.05)。Horne-C)stberg生理时钟问卷调查结果显示:高性能战斗机飞行员PE船的3种基因型在睡眠分型上没有差别。结论高性能战斗机飞行员与普通汉族人PER3基因型频率分布相似。飞行员睡眠分型中,未提示PER3^5与昼行睡眠偏好相关。Objective To identify the genotype frequency of PERIOD3 (PER3) of high performance fighter pilots, and to analyze the relationshipbetween the PER3, genotype and the sleep type. Methods One hundred and four pilots were sampled 2 ml peripheral blood. Genomic DNA was extracted from leucocytes. Genotyping was performed by using polymerase chain reaction. Sleep style of pilots was identified by the Horne-Ostberg questionnaire. The PER3 genotype frequency of the Hart people and high performance fighter pilots was compared. Results The distribution of PER3 genotype frequency of 104: pilots was: PER3^4/4 took 70. 2% (73/104), PER34^4/5 took 26.9% (28/104) and PER3^5/5 took 2. 9% (3/104). Statistic analysis showed that there was no difference between the Han people and pilots on PER3 frequency (X^2= 2. 737, P〉0. 05). Analysis of Horne- Ostberg questionnaire showed'that there was no difference among the three genotypes of high performance fighter pilots. Conclusions What the role of PER35 genotype frequency plays in sleep regulation needs to be further investigated.
