摘  要:为了解山东泰安市猪戊型肝炎病毒(Hepatitis E virus,HEV)感染情况及其对职业人群的危害,从山东省某屠宰场共收集来自泰安市的106份猪胆汁样品,利用逆转录套式聚合酶链式反应(reverse transcription nested polymerase chain reaction,RT-nPCR)检测HEV开放阅读框(open reading frame 2,ORF2)保守区基因,其中32份为阳性,阳性率为30.2%。同时随机挑选20个阳性样品克隆测序,并与GenBank上的参考序列进行同源性比较,发现所获得的序列与9株HEV基因Ⅳ型参考序列之间的核苷酸同源性最高,为83.6%~92.8%。其中3株与中国猪源HEV swGX32株(EU366959)亲缘关系近(90.8%),属于4a亚型,另外17株与新疆猪源HEVswCH25株(AY594199)和人源HEVT1株(AJ272108)亲缘关系近(87.9%~92.8%),属于4d亚型。调查结果表明该地区猪戊型肝炎病毒的阳性率很高,而且进一步证实猪源HEV和人源HEV有较高的同源性,对人类的身体健康具有潜在威胁。To investigate Hepatitis E virus(HEV) infection in swine in Tai'an and evaluate its danger to the working population,106 porcine bile samples from an abattoir were tested for the presence of HEV RNA by reverse transcription nested polymerase chain reaction(RT-nPCR).Thirty-two of 106 samples were positive for HEV RNA,and the positive rate was 30.2%.The nucleotide sequences of 348 bp region within open reading frame 2(ORF2) of 20 swine HEV isolates were determined.The DNAstar software was used for nucleotide sequences analysis.Sequence alignment showed that 20 swine HEV isolates shared nucleotide sequence identities of 83.6%~92.8% with HEV genotypes Ⅳ.Moreover,three of the 20 HEV isolates ??had a 90.8% identity with Chinese swine HEV swGX32(EU366959)belonging to subgenotype 4a,and the other 17 isolates were grouped into subgenotype 4d with a 87.9%~92.8% identity with Xinjiang swine HEV swCH25(AY594199) and Chinese human HEV T1(AJ272108).The results demonstrate that swine is a big reservoir for HEV in Tai'an,and further support the hypothesis that hepatitis E is zoonosis.
