摘  要:目的建立HPLC同时测定3种沙棘叶中3种黄酮苷元(槲皮素、山奈素、异鼠李素)含量的方法。方法采用WatersC18(150.0mm×4.6mm,5μm)色谱柱,以甲醇-乙腈-0.4%磷酸水为流动相进行梯度洗脱,检测波长360nm,柱温25℃。结果在所选的色谱条件下,上述成分在20min内获得较好的分离;3种成分的浓度与峰面积呈良好的线性关系,r>0.9995;样品回收率为93.89%~103.90%,RSD为1.9%~3.2%。测得中国沙棘、辽阜沙棘、野沙棘的黄酮苷含量分别为9.56、17.33、12.37mg/g。结论本方法简单、准确,重现性好,为沙棘中黄酮类成分的测定和质量控制提供依据。Objective To develop a method for simultaneously determinating quercetin,kaempferide,isorhamnetin in Fructus Hippophae by RP-HPLC.Methods The sample was separated on Waters C18 column(150.0 mm×4.6 mm,5 μm),with methanol-acetonitrile-0.4% phosphoric acid solution as the mobile phase.The detection wavelength was set at 360 nm and the column temperature was 25℃.Results The three components were well separated in 20 min.There were good linear relationships between peak area ratio on the mass concentration and correlation coefficient above 0.999 5.The average recoveries and the RSD were 93.89%-103.90% and 1.9%-3.2%,respectively.The flavonoid contents of three different Fructus Hippophae were 9.56,17.33,12.37 mg/g respectively.Conclusion This method is simple and accurate to determinate flavonoid glycosides in Fructus Hippophae.It can be used as an effective way for the quality control and assessment of Fructus Hippophae.
