远志毛状根诱导及其3,6’-二芥子酰基蔗糖酯测定

摘  要:目的从远志诱导出毛状根,并检测其寡糖酯类成分的产生。方法应用发根农杆菌A4、R1600和R1000菌株分别感染远志外植体,采用高效液相色谱(HPLC)法检测诱导出的毛状根中3,6’-二芥子酰基蔗糖酯的含量。结果 3种发根农杆菌均能诱导远志外植体产生毛状根,且毛状根中都检测到3,6’-二芥子酰基蔗糖酯,A4菌株诱导远志下胚轴产生的毛状根的3,6’-二芥子酰基蔗糖酯含量高于R1600和R1000菌株(F=5 066.792,P<0.05)。结论用远志毛状根培养物生产寡糖酯类成分是一条值得探索的新途径。Objective To detect the content of 3,6'-di-O-sinapoyl-sucrose in cultured hairy root induced by Polygala tenuifolia. Methods Hairy roots of Polygala tenuifolia were induced from hypocotyl and cotyledon explants with Agrobacterum rhizogens A4,R1600 and R1000,and their 3,6'-di-O-sinapoyl-sucrose contents were determined by HPLC. Results Hairy roots were all produced from the explants of Polygala tenuifolia,and 3,6'-di-O-sinapoyl-sucrose could be detected in the hairy roots,the levels of 3,6'-di-O-sinapoyl-sucrose produced from rhizogenes induced by A4 were higher than that induced by R1600 and R1000(F=5 066.792,P〈0.05). Conclusion Production of sucrose esters by using hairy root culture of Polygala tenuifolia is a new way worth for further investigation.
