超前镇痛对大鼠炎症痛血小板CD62p水平的影响

摘　　要：目的:探讨氟比洛芬酯用于超前镇痛是否影响血小板的活化状态,为其安全用于围术期疼痛治疗提供理论依据。方法:选择Wistar大鼠24只随机分为4组,每组6只。I组(生理盐水对照组)大鼠右后脚趾部皮下注射5%福尔马林溶液0.1ml之前10min腹腔注射生理盐水溶液2m1。II组(即刻镇痛组)大鼠右后脚趾部皮下注射5%福尔马林溶液0.1ml即刻腹腔注射氟比洛芬酯50mg/kg溶液2ml。Ⅲ、Ⅳ组(超前镇痛低、高剂量组)致痛之前10min腹腔注射25、50mg/kg氟比洛芬酯2m1。致痛后立刻进行疼痛评分;并且分别于致痛前24h、后12h取静脉血检测血小板CD62p水平。结果:预注氟比洛芬酯后有效抑制了相痛反应(P<0.05),对I相无影响(P>0.05)。与I组比较,致痛后12h血小板CD62p水平、Ⅲ、Ⅳ组均降低(P<0.05),且Ⅳ组低于Ⅲ组(P<0.05)。结论:氟比洛芬酯可预防血栓栓塞性疾病。Objective ：To observe the Preemptive analgesic effect of flurbiprofen axetil on the CD62p level of platelet in the inflammatory painful model formalin-induced rats.Methods：Rats were randomized to either I of Ⅳ groups（n=6 rats for each group）：Group I received a intraperitoneal injection of 2 ml of normal saline solution 10min prior to intraplantar injection of 0.1 m1 of 5% formalin solution;Group Ⅱ received a intraperitoneal injection of 2 ml of 50mg/kg flurbiprofen axetil while intraplantar injection of 0.1 m1 of 5% formalin solution,Group Ⅲ、Ⅳ received a intraperitoneal injection of 25、50mg/kg 2 ml of flurbiprofen axetil respectively prior to intraplantar injection of 0.1 m1 of 5% formalin solution.According to Pain reaction weighting score of Dubbisson and Dennis,the accumulative pain score of 0～5min（I phase） and 15～30 min（phase） after formalin injection were recorded respectively.The levels of platelet CD62p were measured respectively.Results ：Intraperitoneal injection of flurbiprofen axetil prior to intraplantar injection of 0.1 m1 of 5% formalin solution inhibit the phase reaction of formalin-induced rats dependently（P0.05）,but not had significant influence on the Ⅰ phase（P 0.05）.Compared to group I,the Ievel of platelet CD62p were significantly less in those of group Ⅲ、Ⅳ,while that of group Ⅳ were less than group Ⅲ significantly（P0.05）.Conclusion ：flurbiprofen axetil can prevent the thrombotic diseases through interfering in activation of platelet.
