摘 要:钩端螺旋体属特异性耐热抗原(TR-Ag)免疫家兔后,家兔血清显微镜凝集试验(MAT)和玻片凝集试验(SAT)出现阳性的时间有明显差异。动物免疫后第3天即可出现SAT阳性,第7~10天达高峰,2周后转为阴性。MAT效价在免疫后2周达高峰,4周后开始下降。提取免疫后7~10天家兔血清IgG、IgM及IgA进行MAT和SAT检测,IgM的MAT效价明显高于IgG,IgA则为阴性,而SAT结果仅IgM为阳性。ELISA夹心法的检测结果也与MAT和SAT一致,表明SAT检出的TR-Ag抗体为IgM,提示应用TR-Ag检测病人相应抗体时应采用早期血清。The time difference when the rabbit serum showed positive results in microscope agglutination test (MAT) and slide agglutination test (SAT) was obvious after the animals were immunized with leptospiral gen us-specific TR antigen. The SAT positive response appeared on the third day with it's reached peak on the seventh to tenth day after immunization, and SAT showed negative results two weeks later. The liter of MAT was highest on the fifteenth day and started declining four weeks after immunization. Enzyme-linked immune sorbent assay (ELISA) was applied to determining the concentration change of IgG and IgM in the rabbit serum and the results were similar to those of SAT or MAT. IgG, IgM and IgA in the immunized rabbits on the seventh to tenth days were extracted and applied to SAT or MAT. Onlycould the IgM produce positive response of SAT. MAT liter of the IgM was obviously higher than that of IgG, and the resull of MAT of the IgA was negative. The results in this study showed that the antibody of TR antigen in SAT was of IgM and suggested that the serum in early period should be taken whed TR antigen was applied lo determining antibody of the patients infected with leptospira.
