摘  要:[目的]研究肾气丸对拟老化大鼠内耳组织线粒体DNA(mtDNA)4834bp缺失发生率的影响。[方法]3月龄大鼠经脑干诱发电位(ABR)检测听阈后分对照组、拟老化组、肾气丸组。拟老化组给予5%D-半乳糖颈部皮下注射连续8周,肾气丸组给予5%D-半乳糖颈部皮下注射连续6周后同时给予肾气丸混悬液灌胃2周。对照组、拟老化组最后2周同时给予等量生理盐水灌胃。[结果]对照组大鼠ABR反应阈与拟老化组、肾气丸组之间差异有统计学意义(P<0.05),拟老化组、肾气丸组之间差异无统计学意义(P>0.05)。拟老化组、肾气丸组与对照组大鼠比较,其内耳组织线粒体4834 bp缺失明显增加,且两组间检出率有明显差异(P<0.05)。[结论]肾气丸能改善大鼠老化过程中的内耳组织线粒体4834 bp缺失的发生率,提示其对大鼠耳蜗老化有一定的改善作用。[Objective] To discuss the effect of Shenqiwan on mtDNA4834 deletion in mimetic aging model rat.[Methods] Three months rats were randomly divided into three groups: group A,which were treated with hypodermic 5 % D-galactose for 8 weeks,and group B(treatment group,n = 10) which were treated daily with Shenqiwan for 2 weeks after hypodermic 5 % D-galactose for 6 weeks,group C were given saline only.Auditory brainstem response(ABR) was used to detect the hearing threshold of rats before and after the experiment.The inner ear tissue was harvested and mitoch-ondrial DNA was amplified to identify the 4834 bp deletion mutation by nested primer polymerase chain reaction(nested PCR) technique.[Result] The threshold of ABR in the group A and group B was significantly higher than that in group C.however the difference in shift of ABR threshold between group A and group B,group C was no significant difference.The mtDNA4834 deletion rate in two model groups(group A /group B) was higher than the control group.There was statistically significant difference between the control group and the two model groups on mtDNA4834 deletion in cochlea.[Conclusion] Shenqiwan can reduce the mtDNA4834 deletion in inner ear of mimetic aging model rats,suggesting that it can improve the aging in rat's cochlear.
