牛黄参含药血清对肝干细胞WB-F 344增殖和凋亡的影响

摘　　要：目的研究牛黄参含药血清对大鼠肝干细胞WB-F344增殖及凋亡的影响,探讨益气解毒法诱导肝干细胞的作用机制。方法采用中药血清药理学方法,以不同浓度的牛黄参含药血清(5%、10%、20%)作用于WB-F344,以正常大鼠血清(5%、10%、20%)处理的WB-F344为空白对照组,以单纯培养体系处理的WB-F344为正常对照组;四甲基偶氮唑盐(methyl thiazolyl tetrazolium,MTT)比色法检测细胞的增殖,取药物血清的最佳增殖浓度,流式细胞仪检测细胞凋亡率。结果经牛黄参含药血清干预后,MTT检测结果显示10%、20%浓度的牛黄参含药血清均能促进WB-F344的增殖(P<0.01),且10%为增殖的最佳浓度梯度;10%牛黄参含药血清作用的WB-F344细胞凋亡率与空白、正常对照组相比显著下降(P<0.01),且以10%牛黄参含药血清成分组效果最佳。结论牛黄参含药血清可在一定程度上促进WB-F344细胞的增殖,抑制其凋亡,这可能是益气解毒法作用于肝干细胞的机制之一。Objective To study the effects of serum containing NHS on proliferation and apoptosis of rat liver stem cellline WB-F 344 and discuss mechanism of the Yiqijiedu method on hepatic stem cell induction. Methods Using traditional Chinese medicine serum pharmacology method,WB-F 344 cells were treated with serum containing different concentrations of NHS（5%,10%,20%）,different concentrations of serum（5%,10%,20%） of the normal rats（blank control group）,and simple culture system（normal control group）.The methyl thiazolyl tetrazolium（MTT） method was used to detect the cell proliferation.The flow cytometer was used to measure cell apoptosis based on the concentration of drug serum for best proliferation. Results Serum containing 10% and 20% NHS could promote the proliferation of WB-F 344（P0.01）,10% was the best.The WB-F 344 cell apoptosis rate intervened by 10% NHS was significantly decreased,compared with the blank and normal control groups（P0.01）,and the ingredient group of serum containing 10% NHS was the best. Conclusion Serum containing NHS can promote the proliferation and inhibit the apoptosis of WB-F 344 cells in a certain extent,which may be one mechanism of the Yiqijiedu method on hepatic stem cells.
