猪卵泡液提取物制备人抑制素α单抗的研究

摘　　要：目的制备人抑制素α(INHα)亚基多肽单克隆抗体(McAb)。方法以猪卵泡液粗提物为免疫原,免疫BALB/c小鼠,取其脾细胞与NS-1小鼠骨髓瘤细胞融合。杂交瘤细胞用人工合成的人抑制素α亚基37～65片段包被,间接ELISA法筛选。结果获稳定分泌抗人抑制素α亚基37～65片段单抗细胞3株。结论免疫学和免疫组化的结果表明3株细胞有特异性和实用性。ObjectiveTo prepare monoclonal antibody against human INH α(McAb).MethodsBALB/c mice were immuned with the extracted substance of porcine follicular fluid (PFF). Then the spleen cells were fused with mouse myeloma cells NS 1. The hybridoma culture supernants were screened by indirect ELISA in which artificial synthetic human inhibin α subunit 37～65 fragment was used as antigen.ResultsThree hybridoma cell lines against inhibin α subunit have been raised.ConclusionAccording to the immunological and immunohistochemical identification, we concluded that three hybridoma cell lines have high sensitivity and high specificity. These antibodies can be used in immunohistochemistry study of the normal and some tumor tissues in human, as well as in procine and rat. [
